AT-序列结合蛋白1基因对人皮肤癌细胞A431凋亡及侵袭能力的影响

【摘要】:目的研究AT-序列结合蛋白1(STAB1)基因对人皮肤癌细胞A431凋亡及侵袭能力的影响。方法以人皮肤癌细胞A431为研究对象,根据不同处理方法分为空白组(A431组)、对照组(NC-siRNA组)和实验组(STAB1-siRNA组)。用lipofectamine 2000脂质体分别转染无关小RNA(NC-siRNA)和针对STAB1的小干扰RNA(STAB1-siRNA)后,计数A431细胞、NC-siRNA A431和STAB1-siRNA A431细胞的增殖情况、细胞凋亡比例和3组细胞的迁移能力。结果从48 h开始,小干扰RNA下调STAB1基因后,A431细胞增殖能力显著降低(P0.05);STAB1-siRNA细胞凋亡比例明显增高(P0.05);A431、NC-siRNA A431及STAB1-siRNA A431细胞划痕后0,48 h在下调STAB1基因后,皮肤癌细胞迁移能力显著降低(P0.05)。结论小干扰RNA下调STAB1基因表达后可促进人皮肤癌细胞A431凋亡,抑制细胞增殖和迁移能力。
[Abstract]:Objective to study the effect of AT- sequence binding protein 1 (STAB1) gene on apoptosis and invasion of human skin cancer cell A431. Methods Human skin cancer cell A431 was divided into blank group (A431 group), control group (NC-siRNA group) and experimental group (STAB1-siRNA group) according to different treatment methods. After transfection of small RNA (NC-siRNA) and small interfering RNA (STAB1-siRNA) targeting STAB1 with lipofectamine 2000 liposome, the proliferation of A431 cells, NC-siRNA A431 and STAB1-siRNA A431 cells, the percentage of apoptosis and the migration ability of the three groups were counted. Results after down-regulation of STAB1 gene by small interfering RNA for 48 h, the proliferation ability of A431 cells decreased significantly (P0.05), the percentage of apoptosis of STAB1-siRNA cells increased significantly (P0.05), and the migration ability of skin cancer cells decreased significantly (P0.05) at 048 h after STAB1 gene was down-regulated by A431 NC-siRNA A431 and STAB1-siRNA A431 cells. Conclusion small interfering RNA can promote the apoptosis of human skin cancer cell line A431 and inhibit cell proliferation and migration after down-regulation of STAB1 gene expression.
