金雀异黄素对UVB氧化损伤HaCaT细胞的保护作用

【摘要】:目的研究金雀异黄素(genistein,Gen)对中波紫外线(UVB)照射人角质形成细胞系HaCaT细胞所致氧化损伤的保护作用。方法体外培养HaCaT细胞,分成空白对照组(A组),25μmol/L金雀异黄素处理组(B组),UVB(30mJ/cm2)辐射组(C组),UVB辐射(30mJ/cm2)+25μmol/L金雀异黄素处理组(D组),MTT法检测细胞增殖,流式细胞仪检测活性氧(ROS)水平,生物化学法测定细胞超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)活性。结果 D组与C组相比,细胞存活率明显增加(C组62%,D组85%),细胞内ROS水平明显降低(C组237.21,D组120.57,抑制率为49%),细胞内SOD和GSH-Px水平明显升高[SOD:C组(180.55±8.34)nU/mL,D组(233.42±6.52)nU/mL;GSH-Px:C组(310.72±22.52)U,D组(380.98±29.50)U],两组之间的差别均有显著的统计学意义。结论 Gen能增加细胞在UVB照射后的存活率,增加细胞内的抗氧化酶活性,降低细胞内ROS水平,对UVB照射所致人角质形成细胞氧化损伤具有保护作用。
[Abstract]:Objective to study the protective effect of genistein (genistein,Gen) on oxidative damage induced by ultraviolet B (UV) (UVB) irradiation on human keratinocytes HaCaT cells. Methods HaCaT cells were cultured in vitro and divided into two groups: blank control group (group A), 25 渭 mol/L genistein treatment group (group B,), UVB (30mJ/cm2) radiation group (group C,), UVB radiation), 25 渭 mol/L genistein treatment group (group D). Cell proliferation was detected by MTT, reactive oxygen (ROS) was detected by flow cytometry, and (SOD), glutathione peroxidase (GSH-Px) activity was measured by biochemical method. Results the cell survival rate in group D was significantly higher than that in group C (85% in group C and 62% in group D), and the level of intracellular ROS in group D was significantly decreased (group C, group 237.21, group D, 120.57, inhibition rate was 49%). Intracellular SOD and GSH-Px levels increased significantly [SOD:C group (180.55 卤8.34) nU/mL,D group (233.42 卤6.52) nU/mL;] GSH-Px:C group (310.72 卤22.52) UD group (380.98 卤29.50) U, there was significant difference between the two groups. Conclusion Gen can increase the survival rate of human keratinocytes after UVB irradiation, increase the activity of antioxidant enzymes, decrease the level of ROS in cells, and protect human keratinocytes from oxidative damage induced by UVB irradiation.
