人类疱疹病毒8型的K14.1亚型与新疆Kaposi肉瘤的相关性研究

【摘要】:目的:明确新疆KS感染HHV-8的K14.1等位基因型,并初步探讨新疆KS不同临床分型及临床表现与不同HHV-8亚型的相关性。方法:使用酚-氯仿-异戊醇法对新疆维吾尔自治区人民医院收集的32例KS石蜡包埋组织进行HHV-8 DNA抽提,并使用巢式PCR扩增K14.1基因片段,确定K14.1等位基因型;最后运用Fisher确切概率法对结果进行统计学分析处理。结果:(1)32例新疆KS中有27例HHV-8感染为阳性,阳性率为84.38%,其中5例AIDS-KS患者HHV-8感染均为阳性,感染率100%;(2)基于K14.1位点,24例为P基因型,3例为M基因型;(3)不同HHV-8 K14.1等位基因型与新疆KS患者粘膜损害率之间的差异性无统计学意义(P0.05);(4)新疆经典型KS和AIDS-KS患者之间不同K14.1等位基因型感染率的差异性无统计学意义(P 0.05)。结论:(1)HHV-8感染在新疆KS的发生发展中起着不可忽视的作用;(2)新疆KS患者感染的HHV-8亚型主要属于K14.1 P等位基因型,也存在部分的K14.1 M等位基因型感染者,其中K14.1等位基因型为国内首次报道;(3)新疆AIDS-KS患者趋向年轻化;(4) HHV-8 K14.1的不同基因型与新疆KS有无黏膜损害无关;(5)新疆KS患者感染的HHV-8 K14.1的不同基因型与KS临床类型无关。
[Abstract]:Objective: to identify the K14.1 allele genotype of HHV-8 infected with KS in Xinjiang and to explore the correlation between different clinical types and clinical manifestations of KS and different HHV-8 subtypes in Xinjiang. Methods: using phenol-chloroform-isoamyl alcohol method, 32 cases of KS paraffin embedded tissues collected from Xinjiang Uygur Autonomous region people's Hospital were extracted by HHV-8 DNA. K14.1 gene fragment was amplified by nested PCR, and K14.1 allele genotype was determined. Finally, the Fisher exact probability method is used to analyze the results statistically. Results: (1) of 32 cases of KS in Xinjiang, 27 cases were positive for HHV-8 infection, the positive rate was 84.38%, among which 5 cases of AIDS-KS patients were all positive for HHV-8 infection, the infection rate was 100%; (2) based on K14.1 locus, 24 cases were P genotype, 3 cases were M genotype, (3) there was no significant difference between different HHV-8 K14.1 allele genotypes and mucosal damage rate of KS patients in Xinjiang (P0.05). (4) there was no significant difference in the infection rate of different K14.1 allele genotypes between Xinjiang classic KS and AIDS-KS patients (P0. 05). Conclusion: (1) HHV-8 infection plays an important role in the occurrence and development of KS in Xinjiang; (2) the HHV-8 subtypes of KS patients in Xinjiang were mainly K14.1P alleles, and there were some K14.1M alleles in Xinjiang. The K14.1 allele genotype was reported for the first time in China. (3) AIDS-KS patients in Xinjiang tend to be younger; (4) the different genotypes of HHV-8 K14.1 are not related to the mucosal damage of KS in Xinjiang; (5) the different genotypes of HHV-8 K14.1 infected by KS patients in Xinjiang are not related to the clinical types of KS.
