人黑色素瘤中TrkB的表达以及雷公藤内酯醇对其表达的影响

【摘要】:目的: 1)研究人黑素瘤组织中TrkB的表达情况,探讨其在黑素瘤中的作用。2)观察TrkB在人黑素瘤细胞A875中的表达情况以及在使用雷公藤内酯醇作用后A875细胞TrkB表达的变化,探讨雷公藤内酯醇抗肿瘤的作用机制。 方法: 1)用免疫组化SP法检测40例人黑素瘤和30例人色素痣组织中TrkB的表达。2)用细胞免疫组化法,RT-PCR法以及western-blot法分别检测雷公藤内酯醇作用前后A875细胞中TrkB的表达情况。 结果: 1)黑素瘤组织和人色素痣组织中TrkB阳性率分别为77.5%和16.7%,两者间差异有统计学意义(P0.01)。 2)a〉在细胞免疫组化法中,TrkB在A875细胞中呈阳性表达,雷公藤内酯醇作用后其表达下降,两者间差异有统计学意义(P0.05)。b〉western-blot法显示雷公藤内酯醇处理A875细胞后TrkB蛋白表达明显低于未处理细胞。c〉RT-PCR法中雷公藤内酯醇作用后TrkB的表达明显低于未使用雷公藤内酯醇作用的A875细胞。 结论: TrkB在人黑素瘤组织中以及人黑素瘤细胞A875中均呈阳性表达,其高表达可能是黑素瘤发生发展的原因之一,在对A875细胞的研究中表明雷公藤内酯醇能抑制TrkB的表达,提示雷公藤内酯醇可能通过抑制TrkB的表达而产生抗肿瘤作用。
[Abstract]:Objective: 1) to study the expression of TrkB in human melanoma. To investigate the role of TrkB in melanoma. 2) to observe the expression of TrkB in human melanoma cell line A875 and the change of TrkB expression in A875 cells treated with triptolide, and to explore the anti-tumor mechanism of triptolide. Methods: 1) immunohistochemical SP method was used to detect the expression of TrkB in 40 cases of human melanoma and 30 cases of human pigmented nevus. 2) the expression of TrkB in 40 cases of human melanoma and 30 cases of human pigmented nevus was detected by immunocytochemistry. RT-PCR and western-blot methods were used to detect the expression of TrkB in A875 cells before and after triptolide treatment. Results: 1) the positive rate of TrkB in melanoma and human pigmented nevus was 77.5% and 16.7%, respectively. There was significant difference between them (P0.01). 2) a > in the cellular immunohistochemical method, the expression of TrkB was positive in A875 cells, and decreased after triptolide treatment. The expression of TrkB protein in A875 cells treated with triptolide was significantly lower than that in untreated cells (P0.05). The expression of TrkB protein in A875 cells treated with triptolide was significantly lower than that in untreated cells (P0.05). The expression of TrkB protein in TrkB treated with triptolide in RT-PCR method was significantly lower than that in untreated cells. The expression was significantly lower than that of A 875 cells treated with triptolide without Tripterygium wilfordii. Conclusion: TrkB is positive in human melanoma tissue and in human melanoma cell line A875, and its high expression may be one of the reasons for the occurrence and development of melanoma. In the study of A875 cell line, triptolide can inhibit the expression of TrkB, and it is suggested that tripterygiolide can inhibit the expression of TNF in human melanoma tissue and human melanoma cell line A875. These results suggest that triptolide may have anti-tumor effect by inhibiting the expression of TrkB.
