老年人视皮质PV和NeuN免疫阳性神经元的形态学观察

【摘要】:目的:研究人脑视皮质中PV、NeuN阳性神经元随着衰老而发生的改变,为研究退行性病变提供形态学依据。 方法:(1)收集22-102岁人脑标本17例。取视皮质,固定,切片,免疫组织化学方法染色视皮质PV、NeuN阳性细胞,光镜下计算细胞密度和光密度值,统计分析。(2)对上述17例视皮质邻片进行6E10免疫组织化学染色,并分析6E10与PV, NeuN的相关性。 结果:1.视皮质内PV阳性神经元主要分布在外颗粒层、外锥体细胞层和内颗粒层。细胞形态大小不一,胞体呈卵圆形,梭形或者不规则形,突起清晰可辨。PV阳性细胞数目随衰老而逐渐下降。2.NeuN阳性神经元分布于视皮质的各层,胞核深染,胞浆和树突淡染。细胞随衰老而出现密度下降、体积变大、空泡化等现象。3.有6E10免疫阳性产物的视皮质,其PV和NeuN免疫阳性神经元数量下降明显。 结论:人视皮质PV和NeuN免疫阳性神经元数目随衰老而下降,有6E10免疫阳性斑块出现的标本,其数目下降更为明显。
[Abstract]:Aim: to study the changes of PVN Neun positive neurons in the visual cortex of human brain with aging, and to provide morphological evidence for the study of degenerative lesions. Methods 17 human brain specimens aged 22 to 102 years were collected. The visual cortex was fixed, sectioned and stained with immunohistochemical staining for PVN positive cells in the visual cortex. The cell density and optical density were calculated under light microscope. Statistical analysis. (2) 6E10 immunohistochemical staining was performed on the adjacent sections of the visual cortex of 17 cases mentioned above, and 6E10 and PV were analyzed. Correlation of NeuN. Results 1. PV-positive neurons in the visual cortex were mainly distributed in the outer granular layer, the outer pyramidal cell layer and the inner granular layer. The size of the cells varied and the cell bodies were oval, fusiform or irregular. The number of clearly identifiable. PV positive cells decreased with aging. 2. Neun positive neurons were distributed in all layers of the visual cortex and the nucleus was deeply stained. Light staining of cytoplasm and dendrites. The density of cells decreased with aging, the volume became larger, vacuolation and other phenomena. 3. There were 6E10 immunoreactive visual cortex. The number of PV and NeuN immunoreactive neurons decreased significantly. Conclusion: the number of PV and NeuN immunoreactive neurons in human visual cortex decreases with aging, and the number of 6E10 immunoreactive plaques decreases more obviously.
