Toll样受体4信号通路在蛛网膜下腔出血后迟发型血管痉挛中的机制研究

【摘要】:目的 本实验拟采用兔蛛网膜下腔出血模型研究Toll样受体4信号通路在蛛网膜下腔出血(SAH)后迟发型脑血管痉挛(DCV)中的作用。 方法 新西兰纯种健康清洁级大白兔60只随机分为3组:①对照组(枕大池注入NS0.5mL/kg),②SAH组③E5564组,每组20只。模型组采用枕大池二次注血法制备蛛网膜下腔出血模型,对照组枕大池内注入生理盐水,E5564组SAH模型建立后,E5564静脉给药。血管造影及TCD评估血管痉挛情况,造模后第7天取材HE染色观察基底动脉痉挛情况和病理变化。免疫组织化学和westernblot方法检测基底动脉Toll受体4(TLR4)和IL-8的的表达。 结果 1.实验结果显示SAH后SAH组基底动脉直径与对照组相比明显变小(P<0.01=; 2.E5564组基底动脉直径与SAH组相比明显减小(P<0.05); 3.E5564组基底动脉TLR4和IL-8的表达与SAH组相比明显减少(P<0.05)。 结论 1.蛛网膜下腔出血(SAH)后迟发型脑血管痉挛(DCVS)可能与Toll样受体4信号通路有关。 2. E5564可以有效的预防SAH后迟发型脑血管痉挛,,部分可能与抑制IL-8的表达有关系。
[Abstract]:Purpose. The purpose of this study was to investigate the role of Toll-like receptor 4 signaling pathway in delayed cerebral vasospasm after subarachnoid hemorrhage in rabbits. Method. Sixty New Zealand purebred healthy clean grade white rabbits were randomly divided into 3 groups: control group (n = 20 in each group, n = 20 in group A, n = 20 in group A, n = 20, n = 20, n = 20 in group A, n = 20). The model of subarachnoid hemorrhage was established in the model group by twice injecting blood into the cistern of the occipital cistern. In the control group, normal saline was injected into the cistern of the occipital cistern and the SAH model was established. The vasospasm was evaluated by angiography and TCD. The vasospasm and pathological changes of basilar artery were observed by HE staining on the 7th day after modeling. The expression of Toll receptor 4 TLR4 and IL-8 in basilar artery were detected by immunohistochemical and westernblot methods. Results. 1. The results showed that the diameter of basilar artery in SAH group was significantly smaller than that in control group after SAH (P < 0.01). 2. The diameter of basilar artery in E5564 group was significantly lower than that in SAH group (P < 0.05). 3. The expression of TLR4 and IL-8 in basilar artery in E5564 group was significantly lower than that in SAH group (P < 0.05). Conclusion. 1. The delayed cerebral vasospasm after subarachnoid hemorrhage may be related to the signal pathway of Toll-like receptor 4. 2. E5564 can effectively prevent delayed cerebral vasospasm after SAH, which may be related to the inhibition of IL-8 expression.
