多发和单发胶质瘤石蜡包埋组织microRNA检测及临床意义的初步探讨

【摘要】:目的: 比较原发性多发胶质瘤和单发性胶质瘤福尔马林固定石蜡包埋(formalin-fixed and paraffin-embedded, FFPE)组织microRNA表达谱的差异,初步探讨显著差异的microRNA及其靶基因在胶质瘤侵袭迁移中可能发挥的作用。 方法: 收集原发性多发胶质瘤石蜡包埋组织标本6例,同时严格按照匹配条件选取单发胶质瘤6例,常规石蜡组织切片,提取总RNA并进行质量检测;采用Agilent公司的人类microRNA芯片进行miRNA表达谱的检测,利用基因芯片分析软件对芯片结果进行数据分析,筛选出多发胶质瘤和单发胶质瘤具有差异表达的microRNA,再利用生物信息学分析软件预测差异表达InicroRNA的靶基因。 结果: 所有FFPE组织标本中提取的总RNA质量检测均符合质控标准。与单发胶质瘤相比,多发胶质瘤中共有128个差异基因有统计学意义(倍数2或者0.5),其中有10个microRNAs显著上调(倍数50):miR-21、miR-455、miR-199、 miR-410、miR-148a、miR-335、miR-34b、miR-664a. miR-542、miR-4453;还有5个表达明显下调(倍数0.05)的InicroRNAs:miR-200a、miR-45、miR-29a、 miR-101、miR-96。基于部分表达差异显著的microRNAs靶基因预测结果,预测到的靶基因涉及到核酸结合、蛋白质结合、转移酶活性、上皮间转化等与肿瘤侵袭迁移相关的多个方面的生物学功能。 结论: 1.原发性多发胶质瘤相比原发性单发胶质瘤存在]microRNA的差异表达,其中miR-21和miR-200a可能与胶质瘤的侵袭和迁移密切相关,有望通过扩大样本进一步深入研究从而寻找到胶质瘤基因诊断和治疗的新靶点; 2. FFPE组织可以满足后续microRNA芯片等实验研究,从而为microRNA生物学功能的研究和探索提供了丰富的资源。
[Abstract]:Objective:. To compare the expression profiles of formalin-fixed formalin-fixed and paraffin-embedded (FFP) in primary multiple gliomas and single gliomas, and to explore the role of microRNA and its target genes in the invasion and migration of gliomas. Methods:. Paraffin-embedded tissue specimens of primary multiple gliomas were collected in 6 cases, and 6 cases of single gliomas were selected strictly according to matching conditions. Routine paraffin sections were used to extract total RNA and detect the quality. The human microRNA chip of Agilent company was used to detect the miRNA expression profile, and the gene chip analysis software was used to analyze the data of the chip. The differentially expressed microRNAs of multiple gliomas and single gliomas were screened and the target genes of differentially expressed InicroRNA were predicted by bioinformatics software. Results:. The quality of total RNA extracted from all FFPE tissue samples was in accordance with the quality control standard. There were 128 differentially expressed genes in multiple gliomas (multiple 2 or 0.5), of which 10 microRNAs were significantly up-regulated (multiple 50: miR-21 miR-455miR-199,miR-410miR-148amiR-338a.miR-542miR-664a. miR-542miR-4453.The other 5 microNASVF miR-200amiR-45miR-29a, miR-101miR-96. MicroRNAs target gene prediction results, The predicted target genes are involved in many biological functions related to tumor invasion and migration, such as nucleic acid binding, protein binding, transferase activity and interepithelial transformation. Conclusion:. 1. The differential expression of microRNA was found in primary multiple gliomas as compared with primary single gliomas, in which miR-21 and miR-200a may be closely related to the invasion and migration of gliomas. It is expected to find a new target for gene diagnosis and treatment of glioma by expanding the sample to further study. 2. FFPE tissue can satisfy the following experimental research such as microRNA chip, which provides abundant resources for the study and exploration of microRNA biological function.
