Tbx3及PTEN在原发性脊髓胶质瘤中的表达及相关性分析

【摘要】:背景与目的: 原发性脊髓胶质瘤约占椎管内肿瘤20%,但在脊髓髓内肿瘤中却占到了80%以上,其主要病理分型包括:室管膜瘤、星形细胞瘤及少突胶质细胞瘤,以室管膜瘤最为常见。目前脊髓胶质瘤的治疗以手术切除为主,并辅以放疗及化疗,但治疗效果都不尽理想,易复发且常出现严重的神经功能障碍的并发症。随着分子生物学的发展,分子靶向治疗和基因治疗为胶质瘤的治疗提供了新的治疗方向,而目前关于脊髓胶质瘤的研究尚不够深入。 PTEN基因是继p53基因后人类发现的又一重要的抑癌基因。其表达的蛋白作为一种肿瘤抑制因子,可以通过阻止过快的细胞生长、分裂速度或者抑制不受控制的细胞分裂方式,进而调控细胞分裂周期。Tbx3是T-box基因家族成员之一,其表达的蛋白具有促进胚胎干细胞分裂增殖的作用。近年来研究的热点认为其与多种恶性肿瘤尤其是乳腺癌的发生密切相关。有研究证实Tbx3的表达可以抑制PTEN的表达从而诱发肿瘤发生发展。 本研究的目的是采用免疫组化的方法回顾性分析Tbx3和PTEN在原发性脊髓胶质瘤中的表达及其相关性,从而进一步从分子生物学角度探讨原发性脊髓胶质瘤的发生机制,为该病的诊治提供新的可行方案。 方法: 收集2010年-2013年确诊为原发性脊髓胶质瘤患者的肿瘤组织石蜡包块共52例,其中室管膜瘤30例、星形细胞瘤12例、少突胶质细胞瘤10例,以病理类型分为3组,,10例正常脊髓组织为对照组。采用免疫组化方法分析各组中Tbx3和PTEN的表达情况,免疫组化结果经图像采集后,采用平均光密度值代表阳性强度进行定量分析。数据结果采用SPSS19.0统计软件进行方差分析各组差异,然后采用Pearson相关性分析法分析Tbx3和PTEN表达的相关性。 结果: Tbx3的各实验组平均光密度值均值均明显高于正常组p0.05,而各实验组间,室管膜瘤组和少突胶质细胞瘤组无明显差异p>0.05,星形细胞瘤组与室管膜瘤及少突胶质细胞瘤组均有明显差异p0.05,且其平均光密度值低于室管膜瘤及少突胶质细胞瘤组。 PTEN的各实验组平均光密度值均值均明显低于正常组,各实验组相对正常组均有明显差异p0.05,且各组平均光密度值均低于正常对照组;而各实验组间,室管膜瘤组和少突胶质细胞瘤组无明显差异p>0.05,星形细胞瘤组与室管膜瘤及少突胶质细胞瘤组均有明显差异p0.05,且其平均光密度值稍高于室管膜瘤及少突胶质细胞瘤组。 在实验组及正常对照组中,PTEN表达与Tbx3表达之间均呈明显负相关r=-0.971,p<0.05,两者之间负相关有统计学意义。 结论: 1.Tbx3在各组脊髓胶质瘤病理类型中均有表达,在正常组中几乎无表达。但在星形细胞瘤中较室管膜瘤及少突胶质细胞瘤中表达较弱,为弱阳性。2.PTEN在各组脊髓胶质瘤病理类型中较正常组均有表达下降。但在星形细胞瘤中较室管膜瘤及少突胶质细胞瘤中表达稍强。3.Tbx3在原发性脊髓胶质瘤中的表达上调,而PTEN的表达下调,两者呈负相关关系。
[Abstract]:Background and Purpose :

The main pathological types of glioma in the spinal cord include : dymodymoma , astrocytic tumor and microglia cell tumor . The main pathological types include : dymodymoma , star - like cell tumor and microglia cell tumor , but the treatment effect is not ideal , and the therapy of molecular targeted therapy and gene therapy provides a new treatment direction for glioma treatment , and the current research on glioma is not enough .

The expression of PTEN gene is one of the members of T - box gene family , and its expressed protein has the function of promoting the division and proliferation of embryonic stem cells . Recent studies have shown that the expression of Tbx3 is closely related to the occurrence of various malignant tumors , especially breast cancer .

The purpose of this study was to investigate the expression of Tbx3 and PTEN in primary glioma by immunohistochemistry .

Method :

The expression of Tbx3 and PTEN was analyzed by means of immunohistochemistry . The expression of Tbx3 and PTEN was analyzed by immunohistochemistry . The results of immunohistochemistry were used to analyze the expression of Tbx3 and PTEN in each group . All groups were analyzed by SPSS 10.0 statistical software . The correlation between Tbx3 and PTEN expression was analyzed by Pearson correlation analysis .

Results :

The mean optical density of Tbx3 group was significantly higher than that in normal group ( p > 0.05 ) , but there was no significant difference between experimental group , dymodymoma group and microglia tumor group ( p > 0.05 ) . The average optical density of Tbx3 group was significantly lower than that in normal group ( p > 0.05 ) .

The mean optical density of PTEN was significantly lower than that of the normal group , and the mean optical density of the experimental group was significantly lower than that of the normal group ( p < 0.05 ) , and the mean optical density of PTEN was lower than that of the normal group .
In the experimental group , there was no significant difference in p > 0.05 , there was a significant difference of p > 0.05 between the group of dymoma and microglia , and the mean value of optical density was slightly higher than that of dymodymoma and microglia cell tumor .

There was a significant negative correlation between the expression of PTEN and the expression of Tbx3 in the experimental group and the normal control group ( r = - 0.971 , p < 0.05 ) .

Conclusion :

1 . The expression of Tbx3 was expressed in the pathological types of glioma in each group , but the expression of PTEN was weak in the normal group .
