β-石竹烯通过抗氧化作用减轻大鼠局灶性脑缺血再灌注损伤

【摘要】:目的:探讨β-石竹烯(β-Caryophyllene,BCP)对脑缺血再灌注(ischemiareperfusion,I-R)损伤大鼠的保护作用及其机制。观察其对核因子相关因子2(2NF-E2-related factor,Nrf2)/血红素氧合酶1(Heme oxygenase-1,HO-1)通路的作用。方法:1.将78只SD大鼠随机分为六组:假手术(Sham)组;模型(I-R)组;β-石竹烯34mg·kg-11组;β-石竹烯102 mg·kg-1组;β-石竹烯306mg·kg-1组;尼莫地平组6mg·kg-1。采用线栓法建立大鼠大脑中动脉栓塞(Middle Cerebral Artery Occlusion,MCAO)模型,缺血1.5h,再灌注24h后,采用双盲法进行神经行为学评分;TTC染色法测定脑梗死体积;透射电镜(Transmission electron microscopy,TEM)和尼氏染色法(Nissl)观察海马神经元病理变化;原位末端凋亡法(TUNEL)检测海马神经元凋亡情况;2.将60只SD大鼠随机分为五组:假手术(Sham)组;模型组(I-R);β-石竹烯34 mg·kg-1组;β-石竹烯102 mg·kg-1组;β-石竹烯306 mg·kg-1组。测定MCAO大鼠海马中超氧化物歧化酶(superoxide dismutase,SOD)、过氧化氢酶(Catalase,CAT)、丙二醛(Malondialdehyde,MDA)、一氧化氮(Nitric oxide,NO)、脂质过氧化物(Lipid peroxidation,LPO)含量;Western法和定量PCR法检测假手术(Sham)组;模型组(I-R);β-石竹烯306 mg·kg-1组Nrf2及HO-1蛋白及基因的表达。结果:1.与Sham组相比,I-R组神经行为学评分升高,脑梗死面积明显增加。β-石竹烯(102mg·kg-1、306 mg·kg-1)与尼莫地平组可不同程度改善MCAO大鼠神经行为学障碍,减少脑梗死面积;同时,β-石竹烯(102mg·kg-1、306 mg·kg-1)与尼莫地平组可减轻海马神经元病理缺损及细胞凋亡情况;2.与Sham组相比,I-R组SOD、CAT活性明显降低,MDA、LPO、NO含量明显增多。与I-R组相比,β-石竹烯(102mg·kg-1、306 mg·kg-1)中,SOD、CAT活性明显增高,MDA、LPO、NO含量明显降低;β-石竹烯(306mg·kg-1)Nrf2、HO-1蛋白及基因的表达明显增多。结论:β-石竹烯对脑缺血再灌注大鼠具有神经保护作用,此作用可能与调控Nrf2/HO-1通路有关。
[Abstract]:Aim: to investigate the protective effect of 尾 -Caryophyllene (尾 -Caryophyllene BCP) on cerebral ischemia-reperfusion injury in rats and its mechanism.To investigate the effect of NRF _ 2 / 1(Heme oxygenase 1(Heme oxygenase-1 (HO-1) pathway on the nuclear factor-related factor 2(2NF-E2-related factor-nrf2 / heme oxygenase 1 (1(Heme oxygenase-1) pathway.Method 1: 1.Seventy-eight SD rats were randomly divided into six groups: sham-operated group; model I-Rgroup; 尾 -caryophylene 34mg kg-11 group; 尾 -caryophyllene 102mg kg-1 group; 尾 -caryophylene 306mg kg-1 group; Nimodipine group 6mg kg-1.The middle Cerebral Artery occlusion model of middle cerebral artery (MCAO) was established by thread occlusion.
