miRNA-21、miRNA-221、miRNA-222在胶质瘤病人血液中的表达及其意义

【摘要】:目的:检测miRNA-21、miRNA-221、miRNA-222在脑胶质瘤病人血液中的表达和探讨miRNA-21、miRNA-221、miRNA-222与脑胶质瘤发生发展的相关性,为脑胶质瘤的早期诊断和进展判断提供了可行的方法和途径。 方法:收集60例术后病理证实为胶质瘤患者的血液标本作为实验组和15例正常人的血液作为对照组。实验组按WHO分级,将I级和II级胶质瘤患者的血液标本归类为低级别胶质瘤组,将III级和IV级胶质瘤患者血液标本归类为高级别胶质瘤组。通过RT-qPCR法分别检测miRNA-21、miRNA-221、miRNA-222在人脑胶质瘤患者血液和正常人血液中的表达量;统计分析miRNA-21、miRNA-221及miRNA-222在实验组与对照组、高级别胶质瘤和低级别胶质瘤组之间的表达差异。 结果: 1.miRNA-21、miRNA-221、miRNA-222在胶质瘤患者血液中的表达水平比正常人血液中的表达均有升高。 2.miRNA-21在高级别胶质瘤患者和低级别胶质瘤患者血液中的表达量对比正常人血液中的miRNA-21表达量均有明显统计学差异(P0.05而且三组之间两两比较表达差异均有统计学学意义)。 3.miRNA-221在高级别胶质瘤血清的表达量与正常人群和低级别胶质瘤患者相比均有统计学差异(P0.05),但在低级别胶质瘤与正常人群相比无统计学差异(P0.05)。 4. miRNA-222仅在高级别胶质瘤组患者血清中表达量与正常人组中的对比表达有统计学差异(P0.05),低级别胶质瘤组与正常人群以及高级别胶质瘤组与低级别胶质瘤组差异较小,无统计学意义(P0.05)。结论: 1.miRNA-21、miRNA-221、miRNA-222在胶质瘤患者的血清中均有较高的表达,提示这三者均可参与胶质瘤的形成。 2.miRNA-21在高级别胶质瘤组和低级别胶质瘤组患者以及正常人组血浆中有差异性表达,提示其可能参与了胶质瘤的形成、发展、恶化过程。 3.miRNA-221、miRNA-222在高级别胶质瘤组血清中表达较高,提示其在高级别胶质瘤形成过程中可能起着重要作用。 4.miRNA-222在高级别胶质瘤组与低级别胶质瘤病人组血液中的表达差异无统计学意义,提示miRNA-221、miRNA-222、miRNA-21表达失衡可能导致胶质瘤的恶化成为高级别胶质瘤。 5.血液中检测miRNA-21、miRNA-221、miRNA-222为胶质瘤的早期无创诊断及其分级和预后评估提供了借鉴意义。
[Abstract]:Objective: to detect the expression of miRNA-21mRNA-221mRNA-222 in the blood of glioma patients and to explore the relationship between miRNA-21mRNA-221 miRNA-222 and the occurrence and development of glioma, which provides a feasible method and approach for the early diagnosis and progression of glioma.Methods: the blood samples of 60 patients with glioma confirmed by pathology were collected as experimental group and 15 normal persons as control group.According to WHO grade, the blood samples of grade I and grade II gliomas were classified as low-grade gliomas, and those of grade III and grade IV gliomas were classified as high-grade gliomas.The expression of miRNA-21mRNA-221mRNA-222 in the blood of human glioma patients and normal people was detected by RT-qPCR method, and the difference of miRNA-21 miRNA-221 and miRNA-222 expression between the experimental group and control group, high grade glioma group and low grade glioma group was analyzed statistically.Results:1. The expression of miRNA-21mRNA-221 miRNA-222 in the blood of glioma patients was higher than that in normal controls.The expression of 2.miRNA-21 in the blood of high grade glioma patients and low grade glioma patients was significantly different compared with that of normal controls (P0.05) and the differences between the three groups were statistically significant.The expression of 3.miRNA-221 in the serum of high grade glioma was significantly different from that of normal and low grade glioma patients (P 0.05), but there was no significant difference between low grade glioma and normal group.4. The expression of miRNA-222 in the serum of patients with high grade glioma was significantly different from that of normal controls (P 0.05), but the difference between low grade glioma group and normal group and between high grade glioma group and low grade glioma group was smaller than that in high grade glioma group and low grade glioma group.There was no statistical significance (P 0.05).Conclusion:1. MiRNA-21mRNA-221 miRNA-222 was highly expressed in the serum of glioma patients, suggesting that all of them were involved in the formation of glioma.The differential expression of 2.miRNA-21 in the plasma of patients with high grade glioma, low grade glioma and normal controls suggests that 2.miRNA-21 may be involved in the formation, development and deterioration of glioma.3. MiRNA-221 miRNA-222 is highly expressed in the serum of high grade gliomas, suggesting that miRNA-222 may play an important role in the formation of high grade gliomas.There was no significant difference in the expression of 4.miRNA-222 between high grade glioma group and low grade glioma patient group, suggesting that the imbalance of miRNA-221 miRNA-222 miRNA-21 might lead to the deterioration of glioma into high grade glioma.5.The detection of miRNA-21mRNA-221 miRNA-222 in blood provides a reference for early noninvasive diagnosis, grading and prognostic evaluation of gliomas.
