ANRIL基因多态性与湖南汉族人群散发颅内动脉瘤的相关性研究

【摘要】:目的:探讨ANRIL基因rs1333040、rs10757272和rs6475606三个单核苷酸多态(SNPs)与湖南汉族人群散发颅内动脉瘤(slA)的相关性。 方法:收集sIA患者108例及对照者107例的临床资料及血液标本,并提取其血液基因组DNA。应用基质辅助激光解吸电离飞行时间质谱(MALDI-TOF-MS)技术对病例组和对照组ANRIL基因的rs1333040、rs10757272和rs6475606三个多态进行基因分型检测。采用多因素二元logistic回归分析比较slA组与对照组、slA组中的单发组和多发组分别与对照组、单发slA组中的破裂组和未破裂组分别与对照组、单发slA组中的前群动脉瘤组和后群动脉瘤组分别与对照组该三个多态基因型及等位基因频率的差异。 结果:(1)slA组和对照组ANRIL基因rs1333040、rs10757272、 rs6475606三个多态基因型及等位基因频率比较均无统计学差异(p0.05,多因素二元logistic回归)。(2)sIA组中的单发slA组和多发slA组ANRIL基因rs1333040、rs10757272、rs6475606三个多态基因型及等位基因频率分别与对照组比较均无统计学差异(p0.05,多因素二元logistic回归)。(3)单发slA组中的破裂sIA组和未破裂slA组ANRIL基因rs1333040、rs10757272、rs6475606三个多态基因型及等位基因频率分别与对照组比较均无统计学差异(p0.05,多因素二元logistic回归)。(4)单发slA组中的前群sIA组和后群slA组ANRIL基因rs1333040、rs10757272和rs6475606三个多态基因型及等位基因频率分别与对照组比较均无统计学差异(p0.05,多因素二元logistic回归)。 结论:(1)ANRIL基因rs1333040、rs10757272和rs6475606三个多态可能与湖南汉族人群sIA的易感不相关。(2)ANRIL基因rs1333040、rs10757272和rs6475606三个多态可能与湖南汉族人群slA单发/多发,单发sIA破裂/未破裂患者发病风险不相关。(3)在湖南汉族单发sIA患者中,ANRIL基因rs1333040、rs10757272和rs6475606三个多态可能与颅内动脉瘤前/后群位置分布无关。
[Abstract]:Objective: to investigate the relationship between the ANRIL gene rs1333040rs10757272 and rs6475606 and the sporadic intracranial aneurysm SNPs in Hunan Han population. Methods: the clinical data and blood samples of 108 patients with sIA and 107 controls were collected and their blood genome was extracted. Matrix assisted laser desorption ionization time of flight mass spectrometry (MALDI-TOF-MS) was used to detect the polymorphism of ANRIL gene rs1333040 rs10757272 and rs6475606 in patients and controls. Multivariate logistic regression analysis was used to compare the single and multiple groups in slA group and control group, and the ruptured group and unruptured group in single slA group and control group respectively. The polymorphism genotype and allele frequency of anterior group aneurysm group and posterior group aneurysm group in single slA group were different from those in control group. Results the ANRIL gene rs1333040rs10757272 was not significantly different among the control group and the control group. There was no significant difference in the frequency of the three polymorphic genotypes and alleles among the three rs6475606 genotypes. There were three polymorphic genotypes of ANRIL gene rs1333040rs10757272rs6475606 in the multivariate logistic regression. There was no significant difference in allelic frequencies between the two groups (p0.05). There was no significant difference in allelic frequencies between the two groups (p0.05, multivariate binary logistic regression analysis) ANRIL gene rs1333040rs10757272rs6475606 in single slA group and unruptured slA group, respectively, as compared with the control group. There was no significant difference in ANRIL gene rs1333040rs10757272 and rs6475606 allele frequencies between pre- and posterior slA groups in multivariate logistic regression analysis and multivariate logistic regression analysis (P 0.05). There was no significant difference in ANRIL genotype and allelic frequencies between the two groups as compared with the control group. P0.05, multivariate logistic regression. Conclusion the three polymorphisms of the ANRIL gene rs1333040rs10757272 and rs6475606 may not be related to the susceptibility of sIA in Hunan Han population. The rs1333040 rs10757272 and rs6475606 polymorphisms of ANRIL gene may be singleton / multiplex with slA in Hunan Han population. The polymorphism of ANRIL gene rs1333040rs10757272 and rs6475606 may not be associated with the distribution of anterior / posterior group of intracranial aneurysms.
