脊髓自噬功能与糖尿病神经病理性疼痛的相关性研究

【摘要】:目的探讨脊髓自噬功能与糖尿病神经病理性疼痛(DNF)的相关性。方法选取STZ制备的糖尿病模型,随机分为制模后3 d组、制模后1周组、制模后2周组、制模后3周组及制模后4周组,每组20只。同期选取5组健康大鼠分为对照1~5组(Con 1~5),每组20只,注射柠檬酸缓冲液。比较各组血糖、机械疼痛阈值、热刺疼痛阈值,以及脊髓自噬功能和脊髓补体C3的表达。结果制模后各组血糖均高于同期对应的Con组(P0.05),除制模后3 d组外,其余各组机械疼痛阈值低于同期对应的Con组(P0.05)。制模后3周组、制模后4周组热刺激疼痛阈值也低于同期对应的Con组(P0.05)。制模后2周组、制模后3周组和制模后4周组C3 mRNA、补体C3、Beclin-Ⅰ和LC3-Ⅱ表达均高于同期对应的Con组(P0.05)。制模后3 d组、制模后1周组、制模后2周组、制模后3周组和制模后4周组P62蛋白表达均低于同期对应的Con组(P0.05)。结论大鼠脊髓补体C3、Beclin-Ⅰ、LC3-Ⅱ、P62激活提示脊髓自噬功能激活;脊髓自噬功能激活可能参与了DNP发生发展。
[Abstract]:Objective to investigate the correlation between spinal cord autophagy and diabetic neuropathic pain (DNF). Methods Diabetic model made by STZ was randomly divided into three groups, one week after model making, two weeks after modeling, three weeks after modeling and four weeks after making model, with 20 rats in each group. At the same time, five groups of healthy rats were divided into control group (n = 20) and control group (n = 20). Blood glucose, mechanical pain threshold, Tottene pain threshold, spinal autophagy and C3 expression in spinal cord were compared. Results the blood glucose in each group was higher than that in Con group at the same time, and the mechanical pain threshold of other groups was lower than that of Con group in the same period except for 3 d after model making. The pain threshold of thermal stimulation in the group of 3 weeks after modeling and 4 weeks after modeling was also lower than that in the corresponding Con group (P 0.05).
