实验性蛛网膜下腔出血后早期铁调素表达的变化

【摘要】:目的:观察蛛网膜下腔出血(subarachnoid hemorrhage,SAH)后早期大鼠海马铁含量及铁调素(Hepcidin)蛋白的表达变化,探讨铁调素参与SAH后早期脑铁代谢紊乱的可能机制。 方法:成年雄性SD大鼠90只,随机分为假手术对照组,实验组(蛛网膜下腔出血组)。采用视交叉前池注血法构造大鼠SAH模型。在SAH后早期12、24、48、72h采用免疫组化、Western-blot法检测大鼠SAH后海马组织内Hepc的蛋白表达情况,使用原子吸收光谱仪对海马组织总铁含量进行检测。 结果:免疫组化实验结果发现,,相对于假手术对照组(0.190.05),大鼠SAH后12h见Hepcidin表达水平明显增高(0.300.06),至72h呈逐渐增高趋势(0.560.07),差异有统计学意义(F=31.911,P㩳0.05);Western blot实验同样发现,与假手术(对照)比较(0.238 0.047),大鼠SAH后12h见Hepcidin蛋白表达水平明显增高(0.481 0.065),至72h呈逐渐增高趋势(1.6270.143),差异有统计学意义(F=147.314,P㩳0.05);原子吸收光谱仪在蛛网膜下腔出血后12h检测到大鼠海马总铁含量开始增加(58.509.19),至72h呈逐渐增高趋势(99.347.68),与假手术对照组比较(43.514.59),差异有统计学意义(F=28.799,P㩳0.05);大鼠海马组织Hepcidin蛋白的表达量与海马铁含量成正相关(r=0.914,P0.05)。结论:大鼠SAH后脑铁含量的增加可能与Hepcidin表达增高有关。
[Abstract]:Aim: to observe the changes of iron content in hippocampus and the expression of iron modulin (hepcidin) protein in hippocampus of rats with subarachnoid hemorrhage (SAH) after subarachnoid hemorrhage (SAH), and to explore the possible mechanism of iron modulin involved in early brain iron metabolism disorder after SAH. Methods: 90 adult male SD rats were randomly divided into sham operation control group and experimental group (subarachnoid hemorrhage group). The rat SAH model was constructed by injecting blood into the anterior cistern of optic chiasma. The expression of Hepc protein in hippocampus of rats with SAH was detected by immunohistochemistry at 12: 24, 48 and 72 hours after SAH, and the total iron content of hippocampus was detected by atomic absorption spectrometer (AAS). Results: compared with the sham-operated control group (0.190.05), the expression of Hepcidin increased significantly 12 hours after SAH, and gradually increased to 0.560.07 at 72 h after SAH. The difference was statistically significant. Compared with sham-operated (control) 0.238 The expression of Hepcidin protein was significantly increased in rats 12 hours after SAH. The content of total iron in hippocampus of rats was detected by atomic absorption spectrometer at 12 hours after subarachnoid hemorrhage, and the total iron content in hippocampus of rats began to increase by 58.509.19 and 99.347.68% at 72 hours, which was significantly higher than that of sham operation control group (P < 0.05). The content of total iron in hippocampus of rats was increased gradually at 72 h after SAH by atomic absorption spectrometer (AAS), and the content of total iron in hippocampus of rats was increased by atomic absorption spectrometer (AAS) at 12 hours after subarachnoid hemorrhage. Compared with 43.514.59, the difference was statistically significant, and the expression of Hepcidin protein in rat hippocampus was positively correlated with the content of iron in hippocampus. Conclusion: the increase of brain iron content after SAH may be related to the increase of Hepcidin expression.
