大鼠坐骨神经损伤背根神经节内P物质和降钙素基因相关肽的表达

【摘要】:目的:神经肽P物质(Substance P SP)、降钙素基因相关肽(Calcitonin gene-related peptide CGRP)共同存在于大鼠坐骨神经损伤后背根神经节中。本研究探讨大鼠坐骨神经损伤后背根神经节内神经肽SP、CGRP阳性表达及意义,今后为进一步探索周围神经损伤后神经肽可能起的作用提供理论支持。方法:收集40只SD(成年雄性大鼠),其中20只SD制备大鼠左侧坐骨神经损伤模型组(A组);10只SD制备大鼠左侧坐骨神经离断后吻合模型组(B组);同时收集SD 10只作为正常手术对照组(C组)。①术后第1w、第2w、第4w:观察A组腰5背根神经节中P物质(SP)、降钙素基因相关肽(CGRP)阳性细胞计数及平均光密度(Mean optical density MOD),并且与C组进行比较。②术后第4w、12w:观察B组腰5背根神经节中SP、CGRP阳性细胞计数及平均光密度(MOD),并且与C组进行比较。③术后第12w:观察B组中的坐骨神经进行组织学观察;观察吻合后坐骨神经的电生理变化;观察B组SP、CGRP神经纤维;蛋白基因产物9.5(protein gene product9.5,PGP9.5)神经纤维。结果:20例大鼠左侧坐骨神经损伤模型组(A组)、10例大鼠左侧坐骨神经离断后吻合模型组(B组)及10例正常对照手术组(C组)均进入结果分析。①术后1 w:A组背根神经节内SP、CGRP的阳性细胞计数及MOD高于C组,两者有显著性差异(p㩳0.05)。②术后2 w:A组背根神经节内SP、CGRP的阳性细胞计数及MOD高于C组两者有显著性差异(p㩳0.05)。③术后4 w:A组背根神经节内SP、CGRP的阳性细胞计数及MOD与C组无明显差异(p0.05)。④术后1w A组背根神经节内SP、CGRP的阳性细胞计数及MOD高于术后2w,两者有显著性差异(p㩳0.05)。⑤A组背根神经节内SP、CGRP的平均光密度在术后第1w达高峰,到第4w恢复正常水平。⑥B组在术后4w背根神经节内SP、CGRP的的阳性细胞计数及MOD低于C组,两者有显著性差异(p㩳0.05)。⑦B组在术后12w背根神经节内SP、CGRP的阳性表达与C组无明显差异(p0.05)。⑧B组在术后第12w SP、CGRP及PGP9.5显示免疫阳性神经纤维,能观察到SP、CGRP神经纤维。B组12w显示坐骨神经功能指标有很好的恢复,说明再生神经中存在SP、CGRP神经纤维。结论:SP、CGRP均不同程度的存在于大鼠坐骨神经损伤后背根神经节中。大鼠坐骨神经损伤后,背根神经节中SP、CGRP的阳性表达在初期显著增加,可以作为早期坐骨神经损伤的一个指标。SP、CGRP不仅参与神经损伤信号传导,而且在大鼠坐骨神经损伤再生修复中可能起调控作用。
[Abstract]:Objective: neuropeptide P (Substance P SP), calcitonin gene related peptide (Calcitonin gene-related peptide CGRP) co exist in the dorsal root ganglion of rat sciatic nerve injury. This study is to explore the positive expression and significance of neuropeptide SP, CGRP in the dorsal root ganglion of rat sciatic nerve injury. 40 SD (adult male rats) were collected, of which 20 SD were used to prepare the rat left sciatic nerve injury model group (group A); 10 SD rats were prepared for the left sciatic nerve anastomosis model group (group B), and 10 were collected as the normal operation control group (group C). (1) the postoperative 1W, 2W and 4w: observed P substance (SP), calcitonin gene related peptide (CGRP) positive cell count and mean light density (Mean optical density MOD) in the 5 dorsal root ganglion of group A, and compared with C group. Histological observation of the sciatic nerve in group B after 12w: was observed, and the electrophysiological changes of the sciatic nerve were observed after the anastomosis; the SP, CGRP nerve fiber, protein gene product 9.5 (protein gene product9.5, PGP9.5) nerve fibers were observed in group B. Results: the left sciatic nerve injury model group (A group) in the left side of the rats, and the left sciatic nerve of the rats in 10 cases were observed. After the postfracture anastomosis model group (group B) and 10 normal controls (group C), the results were analyzed. (1) the number of SP, CGRP positive cells in the dorsal root ganglion and MOD in the 1 w:A groups were significantly higher than those in the C group (P? 0.05). (2) the number of SP in the dorsal root ganglion in group 2 w:A after operation, CGRP positive cell count and greater MOD than those in the group were significantly higher. Difference (P? 0.05). (3) there was no significant difference in the number of SP, CGRP positive cells in the dorsal root ganglion and the MOD and C group in the 4 w:A group (P0.05). (P0.05). There was a significant difference in the count of SP, CGRP positive cells in the dorsal root ganglion and the MOD in the 1W A group after operation (0.05). At the peak, the normal level was restored at 4W. 6. In group B, the number of positive cells in the 4W dorsal root ganglion and the number of CGRP were lower than those in the C group. There was a significant difference between the two groups (P? 0.05). (P?) B group in the dorsal root ganglion of the 12W dorsal root, and there was no significant difference in the positive expression of CGRP. SP, CGRP nerve fiber.B group 12W showed that the function of the sciatic nerve was well restored, indicating the existence of SP, CGRP nerve fibers in the regenerative nerve. Conclusion: SP, CGRP all exist in the dorsal root ganglion of the sciatic nerve injury in rats. After the injury of the sciatic nerve in the rat, the positive of SP and CGRP in the dorsal root ganglion. The expression is significantly increased at the early stage, which can be used as an indicator of the early sciatic nerve injury,.SP. CGRP not only participates in the signal transduction of nerve injury, but also may play a regulatory role in the regeneration and repair of the sciatic nerve injury in rats.
