Fascin-1在颞叶癫痫患者及大鼠匹罗卡品模型脑组织中的表达及意义

【摘要】:目的:肌成束蛋白Fascin-1是一种广泛表达于中枢神经系统的细胞骨架蛋白,,在细胞形态及功能发展中起着重要的作用。许多研究发现Fascin-1与突触重塑有着密切的关系,且Fascin-1的异常调节与许多中枢神经系统(CNS)疾病有关。突触重塑是颞叶癫痫(TLE)重要的病理基础,目前仍不清楚Fascin-1是否也参与到颞叶癫痫的病理过程中呢。因此,本研究旨在研究在颞叶癫痫患者及匹鲁卡品诱导的大鼠模型中Fascin-1蛋白的表达情况,并探讨Fascin-1与颞叶癫痫的关系及意义。 方法:从本课题组已建立的颞叶癫痫患者脑组织库中随机选取20例颞叶癫痫病灶脑组织标本做为病例组进行研究,并收集10例无癫痫病史的脑外伤患者经减压手术后获得的的组织结构正常的颞叶皮质做为对照组;40只雄性SD大鼠被分为对照组(5只)、实验组(35只),实验组大鼠腹腔注射匹罗卡品建立颞叶癫痫模型,在模型点燃后的不同时间点(6h、24h、72h、1W、2W、1M、2M月)断头取脑;分别运用免疫组化、Western blot检测癫痫患者及大鼠模型脑组织中Fascin-1蛋白的动态变化情况;免疫荧光双标被用来对Fascin-1的表达进行定位。 结果:Fascin-1主要在颞叶皮质及海马的神经元细胞胞浆及胞膜中表达,不表达于胶质细胞;且Fascin-1在颞叶癫痫患者中的表达水平明显低于正常对照组(P<0.05)。同样,在癫痫模型大鼠颞叶皮质及海马中,Fascin-1的表达水平在各个时间点都低于对照组(P<0.05,皮质6h时间点除外)。皮质表达最低的时间点是在2M,海马表达最低的时间点是在2W。 结论:Fascin-1蛋白水平在颞叶癫痫患者及匹罗卡品大鼠模型中都被下调,提示Fascin-1蛋白的降低可能与颞叶癫痫的病理过程有着密切关系。
[Abstract]:Aim: myobundle protein (Fascin-1) is a cytoskeletal protein widely expressed in the central nervous system (CNS), which plays an important role in the development of cell morphology and function. Many studies have found that Fascin-1 is closely related to synaptic remodeling, and the abnormal regulation of Fascin-1 is related to many central nervous system (CNS) diseases. Synaptic remodeling is an important pathological basis of temporal lobe epilepsy. It is not clear whether Fascin-1 is also involved in the pathogenesis of temporal lobe epilepsy. Therefore, the purpose of this study was to investigate the expression of Fascin-1 protein in patients with temporal lobe epilepsy and pilocarpine induced rat model, and to explore the relationship between Fascin-1 and temporal lobe epilepsy. Methods: twenty brain tissue specimens of temporal lobe epilepsy were randomly selected from the brain tissue bank of patients with temporal lobe epilepsy. Ten cases of traumatic brain injury without history of epilepsy were collected as control group. 40 male Sprague-Dawley rats were divided into control group (n = 5), experimental group (n = 35) and experimental group (n = 35). Temporal lobe epilepsy model was established by intravenously injection of pilocarpine. At different time points after model kindling, the brain was cut off at different time points (6h / 24h / 72h / 72h / 72W / W / M), and the dynamic changes of Fascin-1 protein in brain tissue of epileptic patients and rats were detected by immunohistochemical Western blot, and the expression of Fascin-1 was detected by immunofluorescence double labeling. Results the expression of Fascin-1 was mainly in the cytoplasm and membrane of neurons in temporal cortex and hippocampus, but not in glial cells, and the expression level of Fascin-1 in patients with temporal lobe epilepsy was significantly lower than that in normal controls (P < 0.05). Similarly, the expression level of Fascin-1 in temporal cortex and hippocampus of epileptic model rats was lower than that of control group at all time points (P < 0.05), except for cortex at 6 h. The lowest expression of cortex was at 2M, and that of hippocampus was at 2W. Conclusion the level of Fascin-1 protein is down-regulated in patients with temporal lobe epilepsy and pilocarpine rat model, suggesting that the decrease of Fascin-1 protein may be closely related to the pathological process of temporal lobe epilepsy.
