维药异黑成熟颗粒对帕金森病模型大鼠氧化应激反应的影响

【摘要】:目的:探讨维药异黑成熟颗粒对帕金森病(Parkinson's disease,PD)模型大鼠行为学及氧化应激反应的影响。方法:采用6-羟基多巴胺(6-OHDA)立体定位单侧双位点毁损法制备PD大鼠模型。设立正常空白对照组,并将造模成功的PD模型大鼠随机分成模型空白对照组、美多巴用药组(美多巴125mg/kg)、维药异黑成熟颗粒低、中、高剂量组(分别为1g/kg、3g/kg、6g/kg),每组10只大鼠,观察给药前后大鼠的行为学变化,给药40天后用免疫组化法检测右侧中脑黑质部酪氨酸羟化酶(TH)细胞表达数量变化及中脑黑质部超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-PX)活性及脂质过氧化氢丙二醛(MDA)含量的变化。结果:行为学方面,维药低剂量组旋转圈数(r/min)为(12.50±1.50),与模型空白对照组比较差异无统计学意义(P0.05),美多巴用药组、维药中、高剂量组旋转圈数(r/min)为(6.00±1.07)、(10.38±1.69)、(7.00±1.31),与模型空白对照组比较差异有统计学意义(P0.01)。TH细胞表达方面,维药低剂量组TH数(个)为(9.13±2.23),与模型空白对照组比较差异无统计学意义(P0.05),美多巴用药组、维药中、高剂量组TH数(个)分别为(50.38±2.33)、(25.36±2.56)、(48.13±4.00),与模型空白对照组比较差异有统计学意义(P0.01)。氧化应激方面,维药低剂量组SOD含量为(111.74±6.22)U/mg、GSH-PX含量为(177.95±4.59) U/mg,MDA含量为(7.02±0.40)nmol/ml,与模型空白对照组比较差异均无统计学意义(P0.05),美多巴用药组、维药中、高剂量组SOD含量为(146.82±6.19).(123.27±5.74).(140.48±11.05)U/mg,GSH-PX含量为(222.92±4.83).(185.46±8.20).(219.03±2.66)U/mg,MDA含量为(4.89±0.24).(6.12±0.29).(5.21±0.38) nmol/ml,与模型空白对照组比较差有统计学意义(P0.01)。维药高剂量组在行为学、TH细胞表达、氧化应激方面,与美多巴用药组比较差异无统计学意义(P0.05),维药中剂量组在行为学、TH细胞表达、氧化应激方面,与美多巴用药组比较差异有统计学意义(P0.05),与维药高剂量组比较差异有统计学意义(P0.01)。结论:维药异黑成熟颗粒高、中剂量可改善PD大鼠的行为学表现,可能具有提高机体抗氧化的能力,其中维药高剂量组作用效果与美多巴用药组相似,优于维药中、低剂量组。
[Abstract]:Aim: to investigate the effect of Yiyimairiao granule on the behavior and oxidative stress of Parkinsonian disease (PDD) model rats with Parkinson's disease (PD). Methods: PD rat model was established by 6-hydroxydopamine 6-OHDAs stereotactic method. Normal blank control group was set up, and the successful PD model rats were randomly divided into model blank control group, medopa drug group (medopa 125 mg / kg 路kg ~ (-1), medium and high dose group (1 g / kg ~ (-3) g / kg ~ (-1) mg / kg ~ (-1) mg 路kg ~ (-1), 10 rats in each group, and 10 rats in each group were treated with medopa (125 mg / kg 路kg ~ (-1) 路kg ~ (-1). The behavioral changes of rats before and after administration were observed. The expression of tyrosine hydroxylase (THH) and the activities of superoxide dismutase (SOD), glutathione peroxidase (GSH-PX) and lipids peroxide in the substantia nigra were detected by immunohistochemical method 40 days after administration. Changes of MDAs in aldehydes. Results: in the behavioral aspect, the number of rotation circles in the low dose group was 12.50 卤1.50g / min, which was not significantly different from that in the model control group (P 0.05). The number of rotation circles in the high dose group was 6.00 卤1.07U / min (10.38 卤1.69) and 7.00 卤1.31g respectively. There was a significant difference in the expression of th between the high dose group and the model control group. The th number in the low dose group was 9.13 卤2.230.There was no significant difference between the low dose group and the model control group (P 0.05), and the expression of th in the Madopa group was higher than that in the model control group, and there was no significant difference in the number of th between the low dose group and the model control group (P 0.05), while in the Madopa group, the number of th was 9.13 卤2.23. The th number in the high dose group was 50.38 卤2.33, 25.36 卤2.56, 48.13 卤4.00, respectively, which was significantly higher than that in the model control group (P 0.01). In terms of oxidative stress, the content of SOD in low dose group was 111.74 卤6.22 U / mg / g GSH-PX (177.95 卤4.59). The content of MDA was 7.02 卤0.40 nmol / ml. There was no significant difference compared with the model control group (P 0.05), and the content of GSH-PX in Madopa group was higher than that in control group. In the high dose group, the content of SOD was 146.82 卤6.19U, 123.27 卤5.74, 140.48 卤11.05 渭 g / g, GSH-PX was 222.92 卤4.83N, 185.46 卤8.20 卤8.20 卤2.66 U / mg MDA, the content of MDA was 4.89 卤0.24.6.12 卤0.29 卤0.29 卤0.21 卤0.38 nmol / ml, which was significantly different from that of the model control group (P < 0.01). There was no significant difference in the expression of th cells and oxidative stress between the high dose group and Madopa group, but there was no significant difference in the expression of th cells and oxidative stress between the medium dose group and the Madopa group. There was a significant difference between Madopa group and Madopa group (P 0.05), and the difference was statistically significant compared with the high dose group (P 0.01). Conclusion: the behavior of PD rats can be improved by the medium dose of Weiyangyimiriao granules, which may improve the ability of anti-oxidation. The effect of high dose group is similar to that of medopa group, which is better than that of medium and low dose group.
