迷走神经刺激对癫痫大鼠海马星形胶质细胞缝隙连接蛋白43表达的影响

【摘要】:目的 观察迷走神经刺激(vagus nerve stimulation,VNS)对海人酸(kainic acid,KA)致癫痫大鼠海马星形胶质细胞缝隙连接蛋白43(connexin43,Cx43)表达的影响,探讨Cx43与癫痫之间的关系及其在VNS治疗癫痫中的作用。 方法 将SD大鼠随机分为对照组、KA组和VNS+KA组,每组10只;KA组和VNS+KA组大鼠左侧脑室注射3μl0.05%KA溶液,制备癫痫模型,对照组注射等量生理盐水;VNS+KA组行VNS。观察各组大鼠行为学变化,记录皮层脑电图(electrocorticogram,ECoG);应用免疫组化染色方法和western blot技术检测各组大鼠海马星形胶质细胞Cx43的表达。 结果 大鼠左侧脑室注射KA3~5min后,KA组大鼠为Racine Ⅳ~Ⅴ级重型癫痫发作,ECoG可见棘波、棘慢波及簇状高尖波等异常脑电发放;VNS+KA组大鼠为Racine I~Ⅲ级轻型发作,脑电发放较KA组减轻,对照组无癫痫发作及异常脑电发放。KA组大鼠海马Cx43阳性细胞数和western blot相对表达量值均明显高于对照组(P<0.01),VNS+KA组大鼠海马Cx43阳性细胞数和western blot相对表达量值明显低于KA组(P<0.01)。 结论 VNS可抑制KA致痫大鼠癫痫发作,,降低癫痫大鼠海马Cx43的高表达。提示癫痫大鼠海马Cx43的表达与癫痫发作有关,降低海马Cx43的表达可能是VNS抑制癫痫发作的机制之一。
[Abstract]:Objective to observe the effect of vagal nerve stimulation (vagus nerve) on the expression of connexin 43 (connexin 43) in hippocampal astrocytes of epileptic rats induced by kainic acid, and to explore the relationship between Cx43 and epilepsy and its role in the treatment of epilepsy. Methods Sprague-Dawley rats were randomly divided into control group (KA) and VNS Ka group (n = 10). Rats in each group were injected with 3 渭 l 0.05 Ka solution in the left ventricle to make epilepsy model. The control group was injected with the same amount of normal saline and the VNS Ka group was given VNSs. The changes of behavior were observed and electrocortical electroencephalogram (ECoG) was recorded and the expression of Cx43 in hippocampal astrocytes was detected by immunohistochemical staining and western blot technique.
