低频重复经颅磁刺激对卒中后抑郁大鼠行为学及海马c-fos、c-jun蛋白表达的影响

【摘要】:目的:探讨重复经颅磁刺激(rTMS)对卒中后抑郁(PSD)模型大鼠行为学及海马c-fos、c-jun蛋白表达的影响。方法:筛选40只健康雄性SD大鼠按随机数字表法分为对照组、假手术组、模型组和治疗组(n=10)。对照组不给予任何干预处理;模型组和治疗组采用大脑中动脉栓塞法(MCAO)和慢性不可预见温和应激法(CUMS)建立PSD模型;假手术组和治疗组给予rTMS治疗;通过蔗糖水消耗实验、旷野实验和水迷宫实验对各组大鼠行为学进行评估;应用免疫组化实验检测各组大鼠海马c-fos、c-jun的阳性细胞灰度值。结果:(1)蔗糖水消耗实验:与模型组比较,治疗组蔗糖水消耗量明显增多(P0.05);(2)旷野实验:治疗组大鼠水平及垂直得分较模型组有显著提高(P0.05);(3)水迷宫实验:治疗组大鼠较模型组学习和记忆能力都有所改善(P0.05);(4)免疫组化实验:治疗组大鼠c-fos阳性细胞灰度值高于模型组(P0.05),各组间c-jun阳性细胞灰度值差异不显著(P0.05)。结论:rTMS能够有效改善PSD模型大鼠的抑郁样行为,其机制可能与海马c-fos、c-jun蛋白的表达有关。
[Abstract]:Aim: to investigate the effects of repeated transcranial magnetic stimulation (rTMS) on the behavior and expression of c-fossil-c-jun protein in hippocampus of poststroke depression (PSD) rats. Methods: forty male Sprague-Dawley rats were randomly divided into control group, sham operation group, model group and treatment group (n = 10). The model group and the treatment group were treated with middle cerebral artery embolization (MCAO) and chronic unpredictable mild stress (CUMS), the sham-operated group and the treatment group were treated with rTMS, and the sucrose water consumption test was used. Field experiment and water maze test were used to evaluate the behavior of rats in each group, and the gray scale of c-fos-c-jun positive cells in hippocampus of each group was detected by immunohistochemistry. Results: (1) Sucrose water consumption experiment: compared with the model group, The consumption of sucrose water in the treatment group was significantly increased (P0.05); (2). The scores of horizontal and vertical in the treatment group were significantly higher than those in the model group (P0.05); (3). The learning and memory abilities of the treatment group were improved compared with the model group (P0.05); (4). Immunohistochemical experiment: the gray value of c-fos positive cells in the treatment group was higher than that in the model group (P0.05), but there was no significant difference in the gray value of c-jun positive cells among the groups (P0.05). ConclusionTwo rTMS can effectively improve the depressive behavior of PSD model rats, and its mechanism may be related to the expression of c-fos-c-jun protein in hippocampus.
