五味子木脂素的抗癫痫作用及神经保护研究

【摘要】:目的:研究分析五味子的有效成分五味子木脂素对大鼠癫痫模型的抗癫痫作用及神经保护。 方法:Wistar大鼠90只,分为三组,对照组10只(不予特殊处理),癫痫模型组40只,五味子木脂素组40只,腹腔注射东莨菪碱硝酸甲酯(1mg/kg),20分钟后给予匹罗卡品(320-350mg/kg,腹腔注射)首先诱导大鼠癫痫持续状态发作,诱导成功后予以地西泮5mg/kg肌肉注射,终止发作。随后1个月(30天)用视频记载大鼠癫痫发作次数及发作过程,1个月内发作3次以上为造模成功。五味子木脂素组给予五味子木脂素80mg/kg喂养,对照组及癫痫组正常大鼠饮食。视频记录三组大鼠的癫痫发作情况。记录完成后,处死并灌注取脑,脑组织按重量体积比1:9加入4℃生理盐水,,制备10%的匀浆。应用分光光度法,考马斯亮兰法定蛋白。检测记录三组大鼠血清、大脑组织中的NO、T-AOC、SOD、MDA含量。分析五味子木脂素对癫痫模型大鼠的神经保护作用。 结果:应用视频记录1个月内上述三组大鼠癫痫发作情况。视频记录结果显示:五味子木脂素组40只鼠有17只出现癫痫发作(发作率为42.5%),癫痫组33只出现癫痫发作(82.5%),对照组0只出现癫痫发作(0%),结果显示五味子木脂素具有明显减少癫痫发作次数的作用。五味子木脂素组大鼠血液及脑组织中SOD和T-AOC活性增高, MDA、NO含量明显降低。 结论: 1.五味子木脂素明显减少癫痫发作次数。 2.五味子木脂素通过减少自由基的产生及抗氧化作用对癫痫脑组织具有保护作用。
[Abstract]:Aim: to study the antiepileptic effect and neuroprotective effect of Schisandra chinensis lignin on rat epileptic model. Methods 90 Wistar rats were divided into three groups: control group (n = 10), epileptic model group (n = 40) and Schisandrin group (n = 40). After 20 minutes of intraperitoneal injection of scopolamine methyl nitrate (1mg/kg), pilocarpine (320-350 mg / kg, intraperitoneal injection) was used to induce epileptic persistent state seizure in rats. After successful induction, diazepam 5mg/kg was injected intramuscularly to terminate the seizure. After 1 month (30 days), the number and process of epileptic seizures in rats were recorded by video, and more than 3 seizures in one month were successfully established. Schisandrin group was fed with Schisandrin 80mg/kg, control group and epileptic group were fed with normal diet. The epileptic seizures were recorded in three groups of rats.
