TACI-Ig融合蛋白对小鼠视神经病理损伤的影响

【摘要】:目的·探讨腹腔注射TACI-Ig融合蛋白对小鼠视神经炎症浸润、髓鞘完整性等的影响。方法·将24只C57BL/6J小鼠随机分为4组,分别为空白对照组、生理盐水注射组、TACI-Ig低剂量注射组(0.4 mg/kg)、TACI-Ig高剂量注射组(4 mg/kg),每组6只。TACI-Ig隔日腹腔注射1次,生理盐水对照组按照同样方式注射生理盐水0.2 mL,空白对照组不给予任何干预。20日后将小鼠麻醉灌注,取眼球和视神经组织,石蜡包埋,切片后行苏木精-伊红(H-E)染色和固蓝(LFB)染色,分别观察炎症细胞浸润情况和髓鞘脱失情况。结果·H-E染色可见空白对照组和生理盐水对照组着色均匀,视神经纤维排列紧密、规则;TACI-Ig低剂量注射组(0.4 mg/kg)视神经结构与两对照组类似;高剂量注射组(4 mg/kg)细胞均匀分布,可观察到炎症细胞少量浸润,炎症细胞浸润评分与其他组比较无明显差异(P=0.610 3)。所有组别视网膜结构清晰、层次分明,神经节细胞单层紧密排列、胞体完整、胞核可见、分布均匀,各组实验动物视网膜各层结构相比较并无明显差异。经LFB染色后光学显微镜下观察,发现视神经髓鞘均无明显脱失和损伤情况,4组之间比较差异无统计学意义(P=0.473 6)。结论·低剂量(0.4 mg/kg)TACI-Ig腹腔注射对视神经、视网膜神经节细胞无损伤,高剂量(4 mg/kg)可能引起视网膜区域炎症少量浸润。
[Abstract]:Objective to investigate the effects of intraperitoneal injection of TACI-Ig fusion protein on inflammatory infiltration and myelin integrity of optic nerve in mice. Methods Twenty-four C57BL / 6J mice were randomly divided into 4 groups: control group, saline injection group (0.4 mg/kg), TACI-Ig high dose injection group (4 mg/kg), 6 mice in each group were intraperitoneally injected with TACI-Ig every other day. The control group was injected with 0.2 mL normal saline in the same way, and the blank control group was not given any intervention. After 20 days, the mice were anesthetized and perfused, the eyeballs and optic nerve tissue were taken and embedded in paraffin. Hematoxylin-eosin (H-E) staining and solid blue (LFB) staining were used to observe the infiltration of inflammatory cells and demyelination of myelin. Results H-E staining showed that the optic nerve fibers in the blank control group and the saline control group were uniform, the optic nerve fibers were arranged closely, the structure of optic nerve in the regular TACI-Ig low-dose injection group (0.4 mg/kg) was similar to that in the two control groups, and the cells in the high-dose injection group (4 mg/kg) were evenly distributed. A small amount of inflammatory cell infiltration was observed, and the score of inflammatory cell infiltration was not significantly different from that of other groups (P0. 6103). In all groups, the retinal structure was clear and stratified, the ganglion cells were closely arranged in a single layer, the cell body was intact, the nucleus was visible and distributed evenly, and there was no significant difference in the structure of each layer of retina among the experimental animals. Under the optical microscope after LFB staining, there was no significant difference in demyelination and injury of optic nerve between the four groups (P0. 4736). Conclusion low dose (0.4 mg/kg) TACI-Ig intraperitoneal injection can not damage the optic nerve and retinal ganglion cells, and high dose (4 mg/kg) may cause a small amount of inflammation infiltration in the retina region.
