Sox17和MBP在未成熟大鼠感染性脑损伤中的表达

【摘要】:目的:了解感染所引起的新生大鼠感染性脑组织中Sox17和(髓鞘碱性磷酸酶)MBP的表达。方法:将64只2日龄SD大鼠随机分为对照组、实验组。实验组生后第2~6 d持续腹腔注射脂多糖(LPS)每日0.6 mg/kg(0.01 rnl/g)。对照组则按照相同的注射方式及注射剂量,给予生理盐水,建立新生大鼠脑白质损伤模型。注射后按观察时间点不同分为1 d、3 d、7 d、15 d,4个亚组(n=8)。处死大鼠取出脑白质,HE染色观察脑白质区病理变化,Western Blot法测定各组脑组织中Sox17和MBP蛋白的表达,RT-RCR法检测Sox17和MBPmRNA的表达变化。结果:实验组大鼠生长发育缓慢,HE染色显示脑白质区出现组织水肿,部分核固缩、核碎裂。PCR和Western Blotting结果 发现Sox17在生后1 d开始表达,7 d达高峰,随后下降,各时间点实验组与对照组相比差异有统计学意义(P0.05)。MBP在实验组的表达显著下降,各时间点实验组与对照组相比差异有统计学意义(P0.05)。结论:生后感染可导致新生大鼠脑白质损伤,MBP表达减少,Sox17表达明显升高,提示Sox17剂量的升高对感染后新生大鼠脑组织可能具有保护性修复作用。
[Abstract]:Aim: to investigate the expression of Sox17 and (myelin alkaline phosphatase) MBP in infective brain tissues of neonatal rats induced by infection. Methods: 64 2-day old SD rats were randomly divided into control group and experimental group. Lipopolysaccharide (LPS) was continuously injected intraperitoneally on the 2nd day after birth of the experimental group for 0.6 mg/kg (0. 01 rnl/g) per day. In the control group, the white matter injury model of neonatal rats was established according to the same injection method and dose. After injection, the rats were divided into 4 subgroups (n = 8) according to different observation time points: 1 day, 3 days, 7 days, 15 days. The expression of Sox17 and MBP in brain tissue was detected by Western Blot method. The expression of Sox17 and MBP mRNA was detected by RT-RCR. Results: the slow growth and development of rats in the experimental group showed edema in the white matter area, partial nuclear contraction, nuclear fragmentation. PCR and Western blotting results showed that the expression of Sox17 reached its peak at 1 day after birth and then decreased. The expression of MBP in experimental group was significantly lower than that in control group at each time point (P0.05), and there was significant difference between experimental group and control group at each time point (P0.05). Conclusion: postnatal infection can reduce the expression of MBP and increase the expression of Sox17 in the white matter of neonatal rats, suggesting that the increase of the dose of Sox17 may have protective effect on the brain tissue of neonatal rats after infection.
