内质网应激在帕金森病发病机制中的研究进展

【摘要】:内质网是真核生物加工、修饰、分泌蛋白质和储存钙离子的重要细胞器。错误折叠/未折叠/突变蛋白在内质网中累积、氧化应激和钙离子平衡紊乱破坏了蛋白质的清除机制,引发内质网应激,因而激活了一种称为未折叠蛋白反应的适应性应激反应。未折叠蛋白反应信号由3种应激感受分子调节,它们诱导独立并汇聚的信号通路来维持内质网稳态,或者在长期应激状态下最终触发细胞死亡。内质网应激在帕金森病的发病进程中有着重要作用,本综述就内质网应激在帕金森病中的发生、发展过程及对帕金森病的影响作一综述。
[Abstract]:Endoplasmic reticulum (ER) is an important organelle for eukaryotic biological processing, modification, secretion of proteins and storage of calcium ions. The accumulation of misfolded / unfolded / mutant proteins in the endoplasmic reticulum (ER), oxidative stress and calcium ion balance disorder destroyed the mechanism of protein clearance and induced endoplasmic reticulum stress. This activates an adaptive stress response called the unfolded protein response. Unfolded protein response signals are regulated by three stress-sensing molecules, which induce independent and convergent signaling pathways to maintain endoplasmic reticulum homeostasis or eventually trigger cell death under long-term stress. Endoplasmic reticulum stress plays an important role in the pathogenesis of Parkinson's disease. This review reviews the occurrence and development of endoplasmic reticulum stress in Parkinson's disease and its effects on Parkinson's disease.
