局灶性脑缺血再灌注后大鼠脑组织bFGF和PDGF-B mRNA表达及其与血管新生关系

【摘要】:目的探讨局灶性脑缺血再灌注(I-R)大鼠脑组织中碱性成纤维细胞生长因子(bFGF))和血小板源性生长因子-B(PDGF-B)的表达及其与血管新生的关系。方法 SD大鼠48只,随机分为假手术组(6只)和大脑中动脉I-R(MCAI-R)组。建立SD大鼠MCAI-R模型。MCAI-R组缺血2h后根据再灌注时间窗的不同分为0h、6h、24h、3d、7d、14d、21d共7组。苏木精-伊红染色后观察脑组织病理变化,原位杂交法检测脑组织bFGF和PDGF-B mRNA的表达,免疫组织化学法检测脑组织CD34蛋白表达并计数微血管密度(MVD)。结果 MCAI-R的6h~21d组脑组织中bFGF mRNA表达与假手术组相比明显升高,差异有显著性(F=41.37,q=3.87~17.30,P0.05);而MCAI-R的24h~14d组与6h组相比,脑组织中bFGF mRNA表达也明显升高,差异有显著性(q=4.31~12.50,P0.05)。MCAI-R的6h~14d组PDGF-B mRNA表达与假手术组相比明显升高,差异有显著性(F=29.22,q=4.00~15.14,P0.05);而MCAI-R的24h~14d组脑组织中PDGF-B mRNA表达与MCAI-R的6h组相比,差异也有显著性(q=3.29~11.14,P0.05)。MCAI-R的24h~14d组脑组织中MVD表达与假手术组相比均明显升高,差异有显著性(F=18.42,q=4.69~11.67,P0.05)。MCAI-R的3d~14d组脑组织中MVD与MCAI-R的6h组相比也明显升高,差异有统计学意义(q=6.06~10.06,P0.05)。且脑组织中bFGF、PDGF-B mRNA表达与MVD呈明显正相关(r=0.725、0.784,P0.01)。结论局灶性脑I-R后脑组织中bFGF与PDGFB mRNA表达升高,并一定程度上促进新生血管形成,这对缺血性脑损伤有重要的保护作用。
[Abstract]:Objective to investigate the expression of basic fibroblast growth factor (bFGF) and platelet-derived growth factor B (PDGF-B) in focal cerebral ischemia-reperfusion (I-R) rats and their relationship with angiogenesis. Methods 48 SD rats were randomly divided into sham-operated group (n = 6) and middle cerebral artery (MCAI-R) group. The MCAI-R model of SD rats was established. The MCAI-R group was divided into 7 groups according to the time window of reperfusion 2 hours after ischemia. The pathological changes of brain tissue were observed after hematoxylin-eosin staining, the expression of bFGF and PDGF-B mRNA in brain tissue was detected by in situ hybridization, the expression of CD34 protein in brain tissue was detected by immunohistochemistry and the microvessel density (MVD).) was counted. Results the expression of bFGF mRNA in 6h~21d group of MCAI-R was significantly higher than that in sham operation group (P 0.05). The expression of bFGF mRNA in brain tissue of MCAI-R 24h~14d group was significantly higher than that of 6 h group, and the difference was significant (P 0.05). The PDGF-B mRNA expression in MCAI-R 6h~14d group was significantly higher than that in sham operation group (P 0.05), and the expression of PDGF-B mRNA in 6h~14d group was significantly higher than that in sham operation group (P < 0.05). The difference was significant (FG 29.22QD 4.00 15.14 P 0.05); There was also a significant difference in the expression of PDGF-B mRNA between the 24h~14d group of MCAI-R and the 6 h group of MCAI-R (qn3.29 11.14 P 0.05). The expression of MVD in the 24h~14d group of MCAI-R was significantly higher than that in the sham operation group (P < 0.05), and the expression of MVD in the brain tissue of the MCAI-R group was significantly higher than that of the sham operation group. The brain tissue MVD in MCAI-R group was significantly higher than that in MCAI-R group (P 0.05). There was a positive correlation between the expression of bFGF,PDGF-B mRNA and MVD in brain tissue (r = 0.725, 0.784, P0.01). Conclusion the expression of bFGF and PDGFB mRNA in the brain tissue after focal brain I-R is increased, and the angiogenesis is promoted to some extent, which plays an important role in the protection of ischemic brain injury.
