Dysbindin在人和实验动物颞叶癫痫灶中的表达及意义

目的:Dysbindin蛋白在中枢神经系统的多个部位广泛表达,它在细胞内囊泡转运、突触囊泡转运和神经递质释放等方面发挥重要的作用。在细胞水平,Dysbindin被认为调节突触前的谷氨酸能传递。大脑中兴奋性的和抑制性的神经递质之间出现平衡失调密切关系到癫痫的发生。而谷氨酸是大脑中含量最为丰富的一种兴奋性神经递质,并且在癫痫活动的启动和扩散中发挥作用。本课题旨在研究Dysbindin在颞叶癫痫患者和氯化锂-匹罗卡品癫痫大鼠模型脑组织中的表达变化,探讨Dysbindin与癫痫发生的关系。方法:在前期从耐药性颞叶癫痫患者中收集的脑组织库中随机选取25例患者的颞叶皮质和10例对照颞叶组织,同时用氯化锂-匹罗卡品诱导建立颞叶癫痫模型,收集大鼠海马组织和邻近皮质。运用免疫荧光和western blot的方法检测Dysbindin表达的部位及变化规律。结果:在人颞叶组织中,Dysbindin在神经元胞浆中表达,其在颞叶癫痫患者中的表达水平较对照组显著增高。同样,Dysbindin在大鼠的实验组和对照组的海马和邻近皮质的神经元中表达。大鼠脑组织的western blotting结果显示:Dysbindi...
