铁蛋白在MPP + 诱导的MES23.5细胞损伤中的保护作用研究

帕金森病(Parkinson’s disease,PD)是一种常见的神经退行性疾病,其特征为黑质致密部(Substantia nigra pars compacta,SNpc)多巴胺(Dopamine,DA)能神经元的的进行性退变,伴随α-突触核蛋白的聚集和路易小体的形成。PD的确切病因至今尚未完全阐明,研究表明PD患者和动物模型黑质(Substantia nigra,SN)铁水平明显增加,且SN铁水平与PD进程相关。脑铁代谢紊乱及铁介导的氧化应激可能是DA能神经元损伤的重要病理机制。铁蛋白是一种中空的球形蛋白质,由两种亚基(H和L)组成,空腔内可储存多达4500个铁原子。铁蛋白H-亚基具有铁氧化酶活性,能将亚铁转化成三价铁。L-亚基存在成核位点,促进铁的矿化和铁核的形成。在体内,铁蛋白有两种形式:去铁铁蛋白(Apoferritin)和含铁铁蛋白(Ferritin)。铁蛋白在维持机体铁稳态中起着重要作用。研究发现PD患者SNpc有大量铁沉积,而铁蛋白水平并没有明显增加。铁含量增加而铁蛋白未出现上调,使SN区DA能神经元更易受到氧化应激损伤。研究已证实星形胶质细胞表达高水平铁蛋白,且可分...
