蛛网膜下腔出血大鼠海马区ROCK2信号通路对自噬的影响

  目的研究蛛网膜下腔出血(Subarachnoid Hemorrhage,SAH)后大鼠海马区ROCK2信号通路对自噬的影响。方法实验用108只无特定病原体级雄性Sprague-Dawley大鼠(350450g),编码后采用随机数字表法等分假手术组(Sham组)、蛛网膜下腔出血组(SAH组)、SAH+ROCK2抑制剂组(Drug组)三个组。Sham组仅颈内动脉分离不刺破血管,SAH模型采用颈内动脉刺破法制作,Drug组在建模成功后给予ROCK2抑制剂(10mg/kg)干预,各组分三个亚组(6h、24h、72h)。穿梭箱试验来观察大鼠各组的逃跑反应次数以及逃跑反应时间,对比各组大鼠学习记忆的改变;HE染色观察各组大鼠海马区神经细胞形状及数量变化;免疫组织化学染色及Western Blot实验分析ROCK2蛋白以及Beclin-1和LC3-Ⅱ在大鼠海马区的表达情况。结果1穿梭箱试验:SAH组各亚组逃跑反应次数减少且逃跑反应时间增多同Sham组对比(P<0.05);Drug组亚组逃跑反应次数增多且逃跑反应时间减少对同SAH组对比(P<0.05)。2 HE染色结...
