沉默SNHG12后BMSCs对脑缺血再灌注损伤的保护作用及机制研究

背景:缺血性脑卒中是由血液供应中断引起的,是世界范围内致死、致残的主要原因。尽管随着新的治疗药物和血管内技术的发展,但全世界每年的缺血性脑卒中发病率和患病率仍在持续上升,缺血性脑卒中后发生的脑组织损伤仍然严重威胁着患者的生存和生活质量。对于缺血性脑卒中后缺血再灌注损伤,目前尚无特效治疗药物,而干细胞移植可能成为最有潜力的治疗方式。既往有研究报道,长链非编码RNA(LncRNA)在调节骨髓间充质干细胞(BMSCs)的功能中起着关键作用,并有研究报道长链非编码RNA小核仁RNA宿主基因12(LncRNA SNHG12)在大鼠脑缺血模型脑组织中表达明显异常。但鲜有报道SNHG12修饰的BMSCs在治疗脑缺血后受损伤的脑组织中所发挥的作用。本研究主要探索沉默SNHG12后BMSCs是否通过激活PI3K/AKT-mTOR信号通路对脑缺血再灌注损伤发挥保护作用及其相应机制。方法:本研究利用氧葡萄糖剥夺/再灌注(OGD/R)处理大鼠脑微血管内皮细胞(BMECs),再用qPCR检测BMECs中不同时间点的SNHG12的表达水平。并利用OGD/R处理的大鼠BMECs与BMSCs或OGD/R预处理的BMS...
