苦参素促进实验性脑梗死小鼠轴突再生及其机制研究

  目的:探讨苦参素(Oxymatrine,OMT)是否可促进小鼠脑梗死后神经功能恢复和轴突再生并探讨其潜在机制。方法:将健康雄性C57BL/6小鼠随机分为假手术组(Sham)、溶剂对照组(Vehicle)和OMT组,采用电凝法制备小鼠局灶性大脑中动脉皮层梗死模型。术后24 h腹腔注射不同剂量OMT溶液或等体积蒸馏水,1次/d,连续14天。术后3d、7d、14d、28d进行Rota rod实验(Rota-rod test)和网格行走实验(Grid walking test),检验OMT促进神经功能恢复的效果并确定最佳剂量。术后14d给各组小鼠健侧相对应皮层注射生物素葡聚糖胺(biotinylated dextran amines,BDA)。术后28d取脑染色,观察各组双侧皮层内被BDA标记神经元胞体和突起的变化情况。术后3d、7d、14d、28d分别取脑组织,采用免疫荧光技术检测各组小鼠梗死灶周围皮层内Gap-43的表达变化,采用免疫蛋白印迹法检测各组小鼠梗死侧脑组织内Gap-43、BDNF及GDNF的表达变化。结果:1.行为学实验结果提示OMT可显著提高脑梗死小鼠的运动功能,降低其行走失...

Gap-43 免疫荧光染色显示,与 sham 组相比,vehicle 组和 OMT 组术后 Gap-43 阳性细胞表达均增多(P < 0.05),提示有轴突再生;与 vehicle组相比,OMT 组 Gap-43 阳性细胞在术后 7 天(P < 0.05)、14 天(P < 0.01)和 28 天(P < 0.05)表达增多,差异有统计学意义,提示 OMT 可持续促进梗死灶周围皮质内轴突再生。
