脐带间充质干细胞及细胞因子共移植治疗脑损伤的分子机理研究

第一章脐带间充质干细胞的分离培养鉴定及体外分化研究背景脐带间充质干细胞(Umbilical cord-derived mesenchymal stem cells,UC-MSCs)主要来自母体妊娠后的脐带,具有强大的细胞分化潜力和细胞增殖能力,其来源丰富,安全,不存在伦理问题且分化潜力较高在临床研究中具有很高的价值。脐带间充质干细胞在体外可被诱导分化为具有神经功能的细胞,在脐带干细胞移植治疗脑卒中等神经损伤疾病的研究中,给予大鼠静脉注射脐带间充质干细胞后,发现大鼠的运动行为明显改善。但脐带间充质干细胞治疗神经损伤类疾病的治疗机理仍不明确。研究目的1.体外获取脐带间充质干细胞并进行流式鉴定。2.利用细胞因子进行脐带间充质干细胞的体外成神经诱导分化并进行鉴定。3.模拟体内环境,构建脐带间充质干细胞体外炎症模型,探讨共移植细胞因子对脐带间充质干细胞的影响。研究方法1.脐带间充质干细胞的分离获取孕鼠脐带组织并使用PBS缓冲液洗净血液,将组织分割后加入胰酶消化并制备单细胞悬液,进行细胞计数。2.脐带间充质干细胞的培养将计数后的细胞,置入含有DMEM/F12的培养基中进行培养并传代。3.脐带间充质...
