烟酰胺腺嘌呤二核苷酸对实验性自身免疫性脑脊髓炎小鼠SIRT3/AMPK/mTOR通路的影响

目的:研究烟酰胺腺嘌呤二核苷酸(Nicotinamide adenine dinucleot-ide,NAD+)对实验性自身免疫性脑脊髓炎(experimental autoimmune encephalomyelitis,EAE)小鼠脑组织中沉默信息调节因子3(Silent information regulator of transcription 3,SIRT3)表达的影响,探究NAD+对EAE小鼠发挥治疗作用可能的通路及机制,为临床多发性硬化的治疗寻找新的思路。方法:1.选用C57BL/6雌性小鼠(周龄8-10周、体重18-20g),以MOG35-55作为免疫抗原,辅以完全弗氏佐剂、结核菌素充分乳化,使结核杆菌H37Ra的终浓度为4mg/ml,于小鼠背部脊柱两旁皮下注射,并分别于免疫的当天(免疫当天为第0天)及第2天,分两次腹腔注射0.5ml百日咳毒素(PTX),构建EAE小鼠模型。2.将实验小鼠随机分为正常对照组(Control组)、EAE模型组、NAD+治疗组,每组8只。免疫后的第1天起...

行石蜡包埋切片后,进行 SIRT3 免疫组织化学染色。结果如图 3A 所E 组与 Control 组相比 SIRT3 阳性细胞表达数目明显减少,而 NAT3 阳性细胞表达数较 EAE 组显著增加。统计分析可得(图 3B):与 Control 组阳性细胞数分别为 2.83±1.169、16.17±4.708,差异具学意义(P<0.01)。NAD+组阳性细胞数为 23.83±4.997,与 EAE 组相异具有统计学意义(P<0.01)。AControl EAE NAD+
