SMAS修饰的PEGDA载电凝胶对MC3T3-E1细胞粘附和增殖的影响

【摘要】:目的:观察甲基丙烯磺酸钠(SMAS)修饰的聚乙二醇双丙烯酸酯(PEGDA)水凝胶对小鼠前成骨细胞(MC3T3-E1细胞)粘附及增殖的影响。方法:通过在PEGDA凝胶前体溶液中加入不同浓度的SMAS小分子,合成出携带不同电量负电荷的新型载电凝胶。扫描电镜观察各组水凝胶的表面结构。检测各组水凝胶的蛋白吸附量。观察种子细胞在各组水凝胶表面的早期粘附,并检测不同时间点下凝胶表面细胞粘附量。用cck8法检测细胞在凝胶表面的增殖情况。结果:随着SMAS浓度的提高,凝胶的蛋白吸附量不断提高,同时随着SMAS修饰量的提高,细胞在凝胶表面得到更好的伸展,同时凝胶表面的细胞粘附数明显提高(P0.05)。而在细胞增殖方面,cck8结果表明随着SMAS修饰量的提高,凝胶表面的细胞增殖也得到明显改善。结论:SMAS小分子修饰的PEGDA凝胶能够有效地促进MC3T3-E1细胞在其表面的粘附和增殖。
[Abstract]:Objective: to observe the effect of PEGDAA hydrogel modified with sodium methallyl sulfonate (SMASS) on mouse preosteoblast MC3T3-E1 cells. Methods: different concentrations of SMAS small molecules were added to the solution of PEGDA gel precursor. The surface structure of hydrogels in each group was observed by scanning electron microscope. The protein adsorption capacity of hydrogels in each group was measured. The early adhesion of seed cells to the surface of hydrogels in each group was observed. . The amount of cell adhesion on the gel surface was measured at different time points. The cell proliferation on the gel surface was detected by cck8 method. Results: with the increase of SMAS concentration, the amount of protein adsorbed on the gel increased. At the same time, with the increase of the amount of SMAS modification, the cells extended better on the gel surface, and the cell adhesion number on the gel surface increased significantly, while the cell proliferation. Cck8 results showed that with the increase of SMAS modification amount. Conclusion the PEGDA gel modified with small molecular weight of PEGDA can effectively promote the adhesion and proliferation of MC3T3-E1 cells on its surface.
