可降解吻合器中云南白药及PLGA的基础研究

【摘要】:目的1.通过实验探索合适的云南白药及PLGA材料配比、选择简单易行膜片制作方法。2.通过实验研究云南白药PLGA混合材料的生物相容性,及其对伤口愈合的影响。 方法第一部分:1.复合材料制备:将云南白药与PLGA粉末材料按不同质量配比(1:5,1:10,1:20)溶解于氯仿中搅拌成糊状,再将其自然平铺于玻璃板上。干燥后裁剪成1.5cm×1.5cm大小复合材料片,干燥冷藏备用。 2.体外降解:将不同配比的混合材料膜片放入模拟肠液内,模拟肠液每星期更换,,并将降解装置放置于37度恒温箱中。于1、2、3、4、6、8周,6个时间点分别取出相应膜片,干燥后计算膜片的失重率,并评价其降解情况。 第二部分:混合材料的生物相容性实验:制备云南白药PLGA混合材料浸提液后进行急性毒性实验、热源实验、溶血实验、微核实验,并通过肌肉内埋置混合材料膜片,行手术切口周围组织HE染色、Masson染色、羟脯氨酸测定等来评价云南白药PLGA混合材料生物相容性,并与橡胶进行对比、讨论其对伤口愈合的影响。 结果1、各质量配比的混合材料均在2-4周降解速率最快,降解约74-78%,6-8周降解放缓。 2、混合材料浸提液急性毒性实验阴性、热源实验体温上升均<0.6℃、溶血率≤5%、微核率≤5‰。 3、动物实验初步表明: ①HE染色中切口周围可见多量炎性细胞浸润,各配比的云南白药与PLGA混合材料组较阳性橡胶对照组均较少; ②炎性反应显示:各配比混合材料组的在同一时间段与橡胶组对比具有显著差异(p<0.05),各配比材料组之间在同一时间段对比有显著差异(p<0.05),各配比材料组在不同时间对比有显著差异(p<0.05); ③Masson染色中切口处胶原纤维为蓝绿色,肌纤维及纤维素为红色。7天时我们可见到炎症细胞堆积,纤维细胞增生,少量胶原沉积,胶原纤细、稀疏。14天时切口处见大量胶原沉积,胶原粗大、密集。 结论1.我们可以通过调节云南白药及PLGA材料之间的配比,使之具有不同的降解速度,挑选适合吻合器的要求的比例。 2.云南白药PLGA混合材料是一种生物相容性良好,且可减少伤口炎性反应、促进伤口愈合的可降解材料。配比为1:5的混合材料组降解率稍快,且溶血率及微核率最低,此配比可能更适用于吻合器的制作。
[Abstract]:Objective 1.The suitable ratio of Yunnan Baiyao and PLGA materials was explored through experiments, and the simple and easy method of making diaphragm was chosen. 2.The biocompatibility of Yunnan Baiyao PLGA mixture and its effect on wound healing were studied experimentally.Method part one: one.Preparation of composite material: Yunnan white medicine and PLGA powder materials were dissolved in chloroform and mixed into chloroform at different mass ratio of 1: 5% 1: 1 1: 20) to form paste, and then they were naturally laid on glass plate.After drying, cut into 1.5cm 脳 1.5cm composite sheet, dry and refrigerate.2.In vitro degradation: the film of mixed material with different ratio is put into the simulated intestinal fluid, which is replaced weekly, and the degradation device is placed in the 37 degree thermostat.The corresponding diaphragm was taken out at 6 times after drying, and the weight loss rate of the diaphragm was calculated after drying and the degradation of the diaphragm was evaluated.The second part: the biocompatibility experiment of the mixture: the acute toxicity test, heat source test, hemolysis test, micronucleus test were carried out after the preparation of the extract of Yunnan Baiyao PLGA mixed material.The biocompatibility of Yunnan Baiyao PLGA mixture was evaluated by HE staining and hydroxyproline determination in the tissues around the incision and compared with rubber to discuss its effect on wound healing.Results 1. The degradation rate of the mixed materials with different mass ratios was the fastest in 2-4 weeks, and the degradation rate slowed down from 6-8 weeks to about 74-78 weeks.2, the acute toxicity test of the extraction solution of the mixed material was negative, the body temperature of the heat source test was less than 0.6 鈩
