植入单纯血管束、感觉神经束的组织工程骨修复大段骨缺损

【摘要】:背景:神经肽作为存在于神经组织的一种内源性活性物质,作用于全身多个系统,调节体内各系统生理功能。目的:观察分别植入单纯血管束、感觉神经束的组织工程骨修复兔大段骨缺损对降钙素基因相关肽、神经肽Y表达的影响。方法:新西兰兔共54只制备股骨大段骨缺损模型,以随机抽签法分为感觉神经束植入组、血管束植入组及对照组,每组各18只,分别给予单纯感觉神经束植入、单纯血管束植入及无血管神经束组织工程骨(对照组)修复治疗,分别于造模后4,8,12周对修复骨段行大体、Masson染色观察,比较修复骨段内降钙素基因相关肽和神经肽Y表达水平。结果与结论:(1)造模后4,8,12周,感觉神经束植入、血管束植入组降钙素基因相关肽和神经肽Y mRNA表达水平均显著高于对照组(P0.05);(2)组织工程骨内降钙素基因相关肽和神经肽Y mRNA表达水平随时间增加先增高后下降,其中第8周降钙素基因相关肽和神经肽Y mRNA表达水平显著高于第4,12周(P0.05);且第4周降钙素基因相关肽和神经肽Y mRNA表达水平显著低于其他时间点(P0.05);(3)组织工程骨内降钙素基因相关肽主要表达于新生骨骨质边缘、骨膜及血管外周;神经肽Y主要表达于髓腔及血管外周;(4)结果说明,骨缺损中血管束或感觉神经束植入均可显著上调神经肽降钙素基因相关肽和神经肽Y合成水平,加快骨质修复进程;但感觉神经束植入组织工程骨可能引起支配区感觉功能丧失,故血管束植入更符合正常人体生理功能需求。
[Abstract]:Background: as an endogenous active substance in nerve tissues, neuropeptides act on multiple systems throughout the body and regulate physiological functions of various systems in vivo.Aim: to investigate the effects of tissue engineering bone grafted with vascular bundle and sensory nerve bundle on the expression of calcitonin gene-related peptide and neuropeptide Y in rabbits with large bone defects.Methods: 54 New Zealand rabbits were randomly divided into three groups: sensory nerve bundle implantation group, vascular bundle implantation group and control group. 18 rabbits in each group were given simple sensory nerve bundle implantation.Simple vascular bundle implantation and tissue engineering bone repair (control group) without vascular and nerve bundles were observed by Masson staining at the 12th week after modeling. The expression levels of calcitonin gene-related peptide (CGRP) and neuropeptide Y (NPY) in the repaired bone segment were compared.Results and conclusion the sensory nerve bundle was implanted in the rat model at 8 ~ 12 weeks after the establishment of the model.The expression of calcitonin gene-related peptide and neuropeptide Y mRNA in vascular bundle implantation group was significantly higher than that in the control group (P 0.05). The expression level of calcitonin gene-related peptide and neuropeptide Y mRNA in tissue engineering bone was increased firstly and then decreased with time.The expression level of calcitonin gene-related peptide and neuropeptide Y mRNA was significantly higher in the 8th week than that in the 4th week, and the expression level of calcitonin gene-related peptide and neuropeptide Y mRNA in the 4th week was significantly lower than that in the other time points (P0.05P0.05).Calcitonin gene-related peptide is mainly expressed at the margin of bone.The results showed that vascular bundle or sensory nerve bundle implantation in bone defect could significantly upregulate the synthesis of neuropeptide calcitonin gene-related peptide and neuropeptide Y.However, sensory nerve bundle implantation may cause loss of sensory function in the innervation area, so vascular bundle implantation is more suitable for physiological function of normal human body.
