医用硅橡胶表面的PEG接枝改性及生物相容性评价

【摘要】:植入青光眼导流装置已成为目前治疗重症青光眼病的主要方法,青光眼导流装置主要采用的是硅橡胶材料,这种材料表面疏水性极强,植入后容易在材料周围形成很厚的纤维包囊,从而使导流装置的功能丧失。本论文用聚乙二醇甲醚丙烯酸酯(PEGMEA)对医用硅橡胶进行了亲水化表面改性,并对其改性原理、表面亲疏水性、抗蛋白粘附性、抗细胞黏附性以及组织生物相容性进行了系统评价。 在三氟甲磺酸的催化作用下,聚(甲基氢硅氧烷)中的部分含Si H基团链段通过链交换反应引入到硅橡胶表面,形成表面功能化硅橡胶。在铂金催化剂的作用下,PEGMEA上的双键与Si H基团发生硅氢加成反应,从而使亲水性聚乙二醇分子链接枝到硅橡胶表面。傅立叶变换衰减全反射红外光谱(ATR-FTIR)和X-射线光电子能谱(XPS)结果显示,通过化学接枝改性可将亲水性PEGMEA分子链引入到硅橡胶表面;水接触角测试结果证实接枝改性后硅橡胶表面的强疏水性得到一定程度的缓解;牛血清白蛋白粘附结果证实,经表面改性后的硅橡胶表面蛋白粘附量降低了约55%;而L929成纤维细胞黏附结果表明,PEGMEA改性硅橡胶上黏附的细胞数量只有原始硅橡胶的65%左右,且细胞基本呈圆球状;细胞毒性评价结果显示,改性后的硅橡胶细胞毒性等级为0级。 通过兔眼部组织植入法进一步评价了亲水化改性硅橡胶的体内组织学反应,通过HE染色切片法对比研究了改性前后硅橡胶植入体的纤维包囊厚度、免疫细胞反应等情况。结果表明,植入2周、4周和6周后,改性硅橡胶周围产生的纤维包囊的厚度均小于未改性的医用硅橡胶,综合三个时间点的纤维包囊厚度统计数据,改性后硅橡胶周围产生的纤维包囊厚度比改性前平均减少约35%左右;但巨噬细胞数量统计结果显示,与原始硅橡胶相比,表面改性硅橡胶并没有明显减少炎症反应。
[Abstract]:Implantation of glaucoma diversion device has become the main method for the treatment of severe glaucoma disease. The glaucoma diversion device mainly uses silicone rubber material, which has strong hydrophobicity on the surface. It is easy to form a thick fiber capsule around the material after implantation, which results in the loss of the function of the diversion device. In this paper, the hydrophilic surface modification of medical silicone rubber was carried out by polyethylene glycol methyl ether acrylate (PEGMEA), and its modification principle, surface hydrophobicity, anti-protein adhesion, anti-cell adhesion and tissue biocompatibility were systematically evaluated. In the presence of trifluoromethyl sulfonic acid (TFOS-catalyzed), some of the Si-containing H groups in the poly (methylhydrosiloxane) were introduced to the surface of silicone rubber by chain exchange reaction to form surface functionalized silicone rubber. Under the action of platinum catalyst, the double bonds on PEGMEA reacted with the Si-H group, thus the hydrophilic polyethylene glycol molecule was linked to the surface of silicone rubber. The results of Fourier transform attenuated total reflectance infrared spectroscopy (ATR-FTIR) and X-ray photoelectron spectroscopy (XPS) show that the hydrophilic PEGMEA molecular chain can be introduced to the surface of silicone rubber by chemical grafting modification. The results of water contact angle test confirmed that the surface hydrophobicity of the modified silicone rubber was alleviated to a certain extent, and the adhesion of bovine serum albumin (BSA) was confirmed. The adhesion amount of surface protein of modified silicone rubber was reduced by about 55%, while the adhesion of L929 fibroblasts showed that the number of cells adhered to the modified silicone rubber was only about 65% of that of the original silicone rubber, and the cells were basically spherical. The results of cytotoxicity evaluation showed that the grade of cytotoxicity of modified silicone rubber was 0. The in vivo histological reaction of hydrophilic modified silicone rubber was further evaluated by rabbit eye tissue implantation method. The thickness of fiber capsule and immune cell reaction of silicone rubber implants before and after modification were compared by HE staining and section method. The results showed that the thickness of fiber cyst around modified silicone rubber was smaller than that of unmodified medical silicone rubber after 2 weeks and 6 weeks of implantation, and the statistical data of fiber cyst thickness at three time points were synthesized. The thickness of fiber capsule around modified silicone rubber decreased by about 35%, but compared with the original silicone rubber, the surface modified silicone rubber did not significantly reduce the inflammatory reaction.
