猪心包衍生医用生物膜材料的制备及生物性能检测

【摘要】:目的:制备新型医用生物膜材料,研究制作方法,评价其生物相容性,为临床做应用准备。 方法:无菌条件下取出随机挑选健康猪的心包,通过胰酶去垢剂、冻融核酸酶两种方法进行脱细胞处理,制备出猪心包衍生医用生物膜。通过光镜观察,与对照组对比,选出脱细胞良好的猪心包衍生医用生物膜,利用大鼠皮下包埋实验对其进行生物相容性检测及吸收时间观察。 结果:(1)胰酶去垢剂法和冻融核酸酶法两组猪心包衍生医用生物膜肉眼观察颜色泛白,膜完整且光滑,质地与新鲜猪心包相比无明显变化。 (2)猪心包衍生医用生物膜光镜下观察可见构成清晰。浆膜层某些区域能看到细胞残留,大部分已脱落。纤维层可见波浪状排列的胶原纤维束。排列无明显紊乱,无溶解现象。光镜下均可见自身细胞基本脱除,新鲜对照组仍有大量固有细胞残留。 (3)大鼠术后未应用抗生素,未感染。切口自然愈合,无红肿及异物排出。大鼠未出现局部、全身系统及行为异常,未出现病危及死亡。 (4)大鼠植入胰酶去垢法获得的猪心包衍生医用生物膜皮下包埋后,一周时没明显变化,二周后开始纤维断裂,四周后淋巴细胞侵入,八周后标本白细胞大量浸润。 结论:(1)胰酶去垢法处理的猪心包衍生医用膜更符合实验要求。 (2)猪心包衍生应用生物膜取材容易、制取方便、价格低廉,有良好的生物相容性,是较理想的可吸收膜替代材料。
[Abstract]:Objective: to prepare a new medical biofilm material, to evaluate its biocompatibility and to prepare for clinical application. Methods: the pericardium of healthy pigs was randomly selected under aseptic condition. The biofilm derived from porcine pericardium was prepared by means of trypsin descaling agent and freeze-thawing nuclease. The biomembrane derived from porcine pericardium was selected by light microscope and compared with the control group. The biocompatibility and absorption time of the biomembrane were detected by rat subcutaneous embedding test. Results 1) the piglet pericarpium derived medical biofilm was white in color, intact and smooth, and had no obvious change in texture compared with fresh porcine pericardium by trypsin defouling method and freeze-thawing nuclease method. 2) the structure of pig pericardium derived medical biofilm can be seen clearly under light microscope. Some areas of the serous layer can see cell residues, most of which have fallen off. A wavy collagenous bundle can be seen in the fibrous layer. There was no obvious disorder and no dissolution in the arrangement. Autologous cells were basically removed under light microscope, and a large number of inherent cells remained in the fresh control group. The rats were not treated with antibiotics and were not infected. The incision healed naturally without redness and foreign body excretion. There were no local, systemic and behavioral abnormalities, and no death and illness. 4) after subcutaneous embedding of porcine pericardial derived medical biofilm obtained by trypsin removing scale method, there was no obvious change at one week, the fibers began to break after two weeks, lymphocytes invaded after four weeks, and a large number of white blood cells were infiltrated in the specimen after eight weeks. Conclusion 1) the porcine pericardial derived medical membrane treated by trypsin decontamination is more suitable for the experiment. 2) the biofilm used in porcine pericarp derivation is an ideal substitute for absorbable membrane because it is easy to obtain, cheap, and has good biocompatibility.
