聚四氟乙烯人工血管对血小板聚集的影响及细胞相容性

【摘要】:背景:聚四氟乙烯人工血管是临床常用的医用材料,但其具体的基础研究内容尚未完善。目的:观察聚四氟乙烯人工血管对血小板聚集的作用及细胞相容性。方法:(1)对血小板聚集的作用:检测聚四氟乙烯人工血管的凝血时间、复钙时间。将生理盐水、蒸馏水及聚四氟乙烯人工血管分别加入兔红细胞悬液中,计算溶血率;(2)细胞生物相容性:将SD大鼠原代心肌细胞分别接种于普通玻璃爬片与聚四氟乙烯人工血管片上,3 d后,检测细胞乳酸脱氢酶释放;5 d后,检测细胞增殖。结果与结论:(1)聚四氟乙烯人工血管对兔血的动态凝血时间、复钙时间没有影响;聚四氟乙烯人工血管的溶血率为2.3%;(2)与接种于普通玻璃爬片上的心肌细胞相比,接种于聚四氟乙烯人工血管片上的心肌细胞增殖及乳酸脱氢酶释放无明显变化;(3)结果表明,聚四氟乙烯人工血管血小板聚集无影响,具有良好的血液相容性与细胞相容性。
[Abstract]:Background: polytetrafluoroethylene (PTFE) prosthesis is a commonly used medical material, but its basic research content is not perfect. Objective: to observe the effect of polytetrafluoroethylene (PTFE) on platelet aggregation and its cytocompatibility. Methods the effect of 1) on platelet aggregation: the coagulation time and calcium recovery time of polytetrafluoroethylene (PTFE) artificial blood vessel were measured. Normal saline, distilled water and polytetrafluoroethylene (PTFE) artificial blood vessels were added to rabbit erythrocyte suspension respectively. Cell biocompatibility: SD rat primary cardiomyocytes were inoculated on glass creeping tablets and polytetrafluoroethylene (PTFE) artificial vascular tablets for 3 days, then the lactate dehydrogenase (LDH) was released for 5 days and cell proliferation was detected. The results showed that PTFE prosthesis had no effect on the dynamic coagulation time and calcium recovery time of rabbit blood, and the hemolysis rate of PTFE artificial blood vessel was 2. 3%) compared with that of cardiomyocytes inoculated on ordinary glass creeping tablets. The results showed that the platelet aggregation of PTFE artificial blood vessel had no effect on the proliferation of cardiomyocytes and the release of lactate dehydrogenase (LDH). The results showed that PTFE artificial blood vessel had good blood compatibility and cytocompatibility.
