乳鼠雪旺细胞对自体颅骨组织生长的影响

【摘要】:目的探讨乳鼠雪旺细胞对自体颅骨组织生长的影响。方法体外培养和纯化乳鼠雪旺细胞,取乳鼠颅骨组织分为对照组和观察组。对照组颅骨组织单独培养,观察组颅骨组织与雪旺细胞共同培养,培养第1、2和3周时免疫组织化学法测定颅骨组织骨钙素和骨桥蛋白水平,酶联免疫法吸附法测定培养液中骨碱性磷酸酶活性含量。结果雪旺细胞在体外被成功培养,纯度89%。不同时间点骨钙素水平有差别(P0.05);观察组和对照组骨钙素水平有差别(P0.05),观察组骨钙素水平比对照组高;观察组和对照组骨钙素水平变化趋势有差别(P0.05)。不同时间点骨桥蛋白水平无差别(P0.05);观察组和对照组骨桥蛋白水平无差别(P0.05);观察组和对照组骨桥蛋白水平变化趋势无差别(P0.05)。不同时间点骨碱性磷酸酶活性有差别(P0.05);观察组和对照组骨碱性磷酸酶活性有差别(P0.05),观察组骨碱性磷酸酶活性比对照组高;观察组和对照组骨碱性磷酸酶活性变化趋势有差别(P0.05)。结论雪旺细胞可以提高颅骨组织骨钙素水平和骨碱性磷酸酶活性,促进颅骨组织的生长。
[Abstract]:Objective to investigate the effect of Schwann cells on the growth of autogenous skull tissue in neonatal rats. Methods Schwann cells were cultured and purified in vitro. The skulls were divided into two groups: control group and observation group. In the control group, the skull tissue was cultured alone, the observation group was co-cultured with Schwann cells, and the levels of osteocalcin and osteopontin in the skull tissue were determined by immunohistochemical method at the 2nd and 3rd week of culture. The activity of bone alkaline phosphatase in culture medium was determined by enzyme-linked immunosorbent assay (Elisa). Results Schwann cells were successfully cultured in vitro and the purity was 89%. The levels of osteocalcin in observation group and control group were different (P0.05), the levels of osteocalcin in observation group were higher than that in control group (P0.05), and the trend of osteocalcin level in observation group and control group was different (P0.05). There was no difference in osteopontin level at different time points (P0.05); there was no difference in osteopontin level between observation group and control group (P0.05); there was no difference between observation group and control group (P0.05). The activity of bone alkaline phosphatase in observation group and control group was different (P0.05), the activity of bone alkaline phosphatase in observation group was higher than that in control group (P0.05), and the change trend of bone alkaline phosphatase activity in observation group and control group was different (P0.05). Conclusion Schwann cells can increase osteocalcin level and bone alkaline phosphatase activity and promote the growth of skull tissue.
