类人胶原蛋白-微孔淀粉的制备及生物学评价

【摘要】:文中以微孔淀粉为主体,研究一种新型复合止血材料的制备及生物学评价。此复合材料由一定比例的α-淀粉酶与糖化酶复合后处理淀粉以形成微孔结构,然后复合类人胶原蛋白,再以戊二醛为交联剂进行交联;用SD大鼠作肝脏出血模型进行止血效果对比试验。通过正交试验确定微孔淀粉最优制备工艺条件如下:α-淀粉酶:糖化酶=1∶3,T/℃=50,t/h=20,pH=4.8;用0.2%(V/V)戊二醛乙醇溶液交联的类人胶原蛋白-微孔淀粉止血材料,在相同的止血时间下,该复合材料平均吸水率为3 580%,对照组(分别是微孔淀粉和类人胶原蛋白海绵)的平均吸水率为3 160%和3 280%。而在SD大鼠肝脏实验中,复合材料t/s=94±13,而外购的止血剂t/s=121±15。类人胶原蛋白-微孔淀粉复合材料止血的止血原理从3个方面展开:①微孔淀粉吸收水分并将血液中的有形物质聚集到表面;②加速内源性凝血因子的激活使其形成血栓;③在类人胶原蛋白的牵引下,淀粉颗粒聚成团大幅度增加了比表面积,使止血效果更优秀;对照组止血机理只包含了该复合材料的前两个方面。结果显示,复合材料的止血效果要优于普通微孔淀粉制成的止血剂,证明该复合止血材料具有良好的止血性能。
