振荡流动对组织工程用灌注式生物反应器中剪切力分布影响
[Abstract]:Aim to investigate the effects of oscillatory flow, pore size and porosity of three-dimensional scaffolds on the distribution of velocity and shear force in bioreactor. Based on the theoretical results, an optimized method for preparation of three-dimensional scaffold and perfusion bioreactor for acellular bone was put forward. Methods the three-dimensional acellular bone scaffolds for bone tissue engineering were simplified into isotropic porous media by means of a three-dimensional scaffold and an perfusion bioreactor, which were prepared earlier in the laboratory. The flow velocity and shear force distribution in the bioreactor were modeled theoretically. Results the velocity and Darcy shear force in porous scaffolds were consistent with each other under oscillatory flow. The difference of velocity and Darcy shear force at different radii was decreased, which was beneficial to uniform three-dimensional culture of seed cells in bone tissue engineering. The mean Darcy shear force could be increased by increasing the velocity of perfusion at the inlet, but the mean Darcy shear force could be significantly decreased by increasing the pore diameter or porosity of the scaffolds, which had little effect on the peak velocity of flow in the scaffolds. Increasing the oscillation frequency of inlet oscillatory flow can reduce the maximum peak velocity and the difference of velocity at different radii. Conclusion the suitable oscillating flow is easy to produce shear force for bone tissue engineering stem cells, and the results may provide theoretical guidance for optimizing the three-dimensional culture method of seed cells in bone tissue engineering.
