新型组织工程化改性胶原-壳聚糖复合支架制备与神经细胞的相容性
[Abstract]:Background: the treatment of peripheral nerve defects is still challenging in regenerative medicine. The application of stent materials is considered to be one of the promising exploration directions. The ideal scaffold materials should be safe, durable and lack of antigenicity. Aim: to study the preparation of a new type of tissue engineering modified collagen chitosan scaffold and explore the possibility of using it as a bridge to repair nerve injury in rat sciatic nerve model. Methods: collagen-chitosan composite scaffolds were prepared, and then the scaffolds were chemically modified with 1-ethyl-3-(3-dimethylaminopropyl)-carbodiimide and N-hydroxysuccinimide. Neural stem cells were then implanted on the scaffold, and then together with the scaffold-seed cells, the sciatic nerve defect was implanted into the body. Results & conclusion: the modified cross-linked collagen-chitosan composite scaffolds have three-dimensional reticular structure. Four weeks after transplantation of scaffold-seeded cells, the expression of neurofilament 200 was found in injured sciatic nerve. 4 weeks after transplantation, the expression of neurofilament 200 was found in the injured sciatic nerve. The results showed that the cross-linked collagen-chitosan composite scaffolds with tissue engineering had good cellular compatibility.
