大鼠骨髓间充质干细胞对不同品系小鼠移植皮肤存活时间的影响及机制

【摘要】:目的探讨经尾静脉注射大鼠骨髓间充质干细胞(BMMSC)对不同品系小鼠移植皮肤存活时间的影响及机制。方法分离SD大鼠BMMSC培养至第3代。用皮肤取样器切取60只ICR乳鼠圆形皮片(直径12 mm),将其移植至成年C57BL/6小鼠背部,建立不同品系小鼠间皮肤移植免疫排斥模型。将60只模型小鼠随机均分为PBS组及低、中、高剂量BMMSC组,分别经尾静脉注射PBS、5×10~4个BMMSC、10×10~4个BMMSC、20×10~4个BMMSC各0.2 m L。记录各组移植皮肤存活时间。术后第7天,HE染色观察移植皮肤病理组织学变化;每组随机取5只小鼠处死取其脾脏组织及血清,分别用RT-PCR及ELISA法检测IL-10、TNF-α、IFN-γ。结果中、高剂量BMMSC组皮肤存活时间较PBS组、低剂量BMMSC组长(P均0.05);中、高剂量BMMSC组皮肤存活时间比较,P0.05。术后第7天,病理切片镜下观察显示PBS组炎性细胞浸润最明显,高剂量BMMSC组炎性细胞浸润最少。术后第7天,与PBS组比较,低、中、高剂量BMMSC组脾脏组织TNF-α、IFN-γmRNA相对表达量低(P均0.05)。与PBS组比较,中、高剂量BMMSC组血清IL-10水平高(P均0.05),低、中、高剂量BMMSC组血清TNF-α、IFN-γ水平低(P均0.05);与低剂量BMMSC组比较,中剂量BMMSC组血清TNF-α、IFN-γ水平低(P均0.05),高剂量BMMSC组血清IL-10水平高、TNF-α水平低(P均0.05)。结论经尾静脉注射适当剂量的大鼠BMMSC可延长不同品系小鼠移植皮肤存活时间;该作用可能与BMMSC调控受体小鼠IL-10、TNF-α、IFN-γ的表达有关。
[Abstract]:Objective to investigate the effect and mechanism of rat bone marrow mesenchymal stem cell (BMMSC) injection via tail vein on survival time of skin graft in different strains of mice. Methods SD rat BMMSC was isolated and cultured for the third passage. 60 ICR milk samples were obtained by skin sampler. Mouse round skin grafts (12 mm in diameter, transplanted to the back of adult C57BL / 6 mice, An immune rejection model of skin graft was established in different strains of mice. 60 mice were randomly divided into PBS group and low, medium and high dose BMMSC groups. 5 脳 10 ~ 4 BMMSC 10 脳 10 ~ 4 BMMSC were injected into the tail vein respectively. The survival time of the skin grafts in each group was recorded. The histopathological changes of the transplanted skin were observed by HE staining on the 7th day after operation, and the spleen tissues and serum of 5 mice in each group were killed at random. IL-10 TNF- 伪 and IFN- 纬 were detected by RT-PCR and ELISA respectively. The results showed that the skin survival time of high dose BMMSC group was higher than that of PBS group, and that of low dose BMMSC group was 0. 05%, while that of high dose BMMSC group was 0. 05%. The pathological examination showed that the infiltration of inflammatory cells was the most obvious in PBS group and the least in high-dose BMMSC group. On the 7th day after operation, compared with PBS group, the relative expression of TNF- 伪 -IFN- 纬 mRNA in spleen tissue of high dose BMMSC group was lower than that of PBS group (P < 0.05), and compared with PBS group, the relative expression of TNF- 伪 -IFN- 纬 mRNA in spleen tissue of high dose BMMSC group was lower than that of PBS group (P < 0.05). The serum IL-10 levels in the middle and high dose BMMSC groups were 0.05 and 0.05, while those in the middle and high dose BMMSC groups were 0.05 and 0.05, respectively, which were compared with those in the low dose BMMSC group, while in the middle and high dose BMMSC groups, the serum levels of TNF- 伪 and IFN- 纬 were lower than those in the low dose BMMSC group. The serum levels of TNF- 伪 IFN- 纬 in the middle dose BMMSC group were 0.05%, while in the high dose BMMSC group the levels of TNF- 伪 and TNF- 伪 were 0.050.Conclusion injection of appropriate dosage of rat BMMSC via caudal vein can prolong the survival time of skin grafts in different strains of mice. This effect may be related to the regulation of the expression of IL-10, TNF- 伪 and IFN- 纬 by BMMSC.
