ILK及PTEN在眼睑结膜鳞状细胞癌组织中的表达及临床意义

【摘要】:目的:研究整合素连接激酶(integrin-linked kinase, ILK)和抑癌基因PTEN(10号染色体上缺失与张力蛋白同源的磷酸酶基因)在眼睑结膜鳞状细胞癌中的表达及其与眼睑结膜鳞状细胞癌的分化程度的关系和相关性,探讨它们在眼睑结膜鳞状细胞癌发生发展中的作用和临床意义。 方法:1、收集手术切除的34例眼睑结膜鳞状细胞癌标本的癌组织、10例相应癌旁正常眼睑结膜组织,经4%多聚甲醛固定后,常规脱水,石蜡包埋。 2、应用免疫组织化学EliVisionTMplus法检测PTEN蛋白和ILK蛋白在眼睑结膜鳞状细胞组织、相应癌旁正常眼睑结膜组织中的表达情况。 结果:1、PTEN蛋白在眼睑结膜鳞状细胞癌组织、癌旁正常眼睑结膜组织中的表达阳性率分别为23.53%(8/34),100.00%(10/10),PTEN蛋白在眼睑结膜鳞状细胞癌组织的表达明显低于癌旁正常眼睑结膜组织,差别有显著性(P=0.000,P0.01)。ILK蛋白在眼睑结膜鳞状细胞癌组织、癌旁正常眼睑结膜组织中的表达阳性率分别为73.53%(25/34),0.00%(0/10),ILK蛋白在眼睑结膜鳞状细胞癌组织的表达明显高于癌旁正常眼睑结膜组织,差别有显著性(P=0.000,P0.01)。 2、PTEN蛋白和ILK蛋白的表达与患者的年龄、性别无显著性差别(P0.05)。ILK在眼睑结膜鳞状细胞癌23例高-中分化组中,有14例表达阳性,在11例低分化组中全部表达阳性,两者间有显著性差别(P=0.017,P0.05)。PTEN在眼睑结膜鳞状细胞癌23例高-中分化组中,有8例表达阳性,在11例低分化组中全部表达阴性,两者间有显著性差别(P=0.034,P0.05)。 3、34例眼睑结膜鳞状细胞癌组织中,PTEN蛋白表达阳性者8例,阴性者26例。ILK蛋白表达阳性者25例,阴性者9例。PTEN蛋白和ILK蛋白表达都呈阳性的3例,都呈阴性的6例;PTEN蛋白表达阴性的26例中,ILK蛋白有19例阳性,ILK蛋白表达阴性的10例中,PTEN蛋白有7例阳性。经Spearman等级相关分析,两者表达具有负相关性(r=-0.394,P=0.021,P0.05)。 结论:1、PTEN(?)白在癌旁正常眼睑结膜组织中高表达,在眼睑结膜鳞状细胞癌中阳性表达率显著减低。PTEN蛋白表达与肿瘤的分化程度相关,提示PTEN蛋白低表达或缺失与眼睑结膜鳞状细胞癌的发生、发展有关。 2、检测ILK在眼睑结膜鳞状细胞癌组织中的表达明显高于癌旁正常眼睑结膜组织,提示ILK蛋白的表达可能与眼睑结膜鳞状细胞癌的发生有关:ILK表达与肿瘤分化程度密切相关,提示ILK可能与眼睑结膜鳞状细胞癌的发展及恶性潜能有关。 3、ILK蛋白的表达上调可能抑制PTEN的活性,PTEN蛋白的表达缺失可能促进ILK蛋白的表达,二者共同参与了眼睑结膜鳞状细胞癌的发生发展。ILK基因有可能成为治疗眼睑结膜鳞状细胞癌的新靶点。
[Abstract]:Objective: to study integrin-linked kinase of integrin-linked kinase. ILK) and tumor suppressor gene PTEN (phosphatase gene homologous to tensin on chromosome 10). The relationship between the expression and the differentiation degree of squamous cell carcinoma of eyelid conjunctiva in eyelid conjunctiva squamous cell carcinoma. To investigate their role and clinical significance in the development of squamous cell carcinoma of eyelid conjunctiva. Methods:. 1. 10 cases of normal eyelid conjunctival conjunctiva conjunctiva tissue adjacent to carcinoma were collected from 34 cases of eyelid conjunctival squamous cell carcinoma. After fixed with 4% paraformaldehyde, conventional dehydration and paraffin embedding were performed. 2Immunohistochemical EliVisionTMplus method was used to detect PTEN protein and ILK protein in squamous cell of eyelid conjunctiva. Expression of normal eyelid conjunctival tissue adjacent to cancer. Results the positive rates of PTEN protein in the conjunctival squamous cell carcinoma of the eyelid and the adjacent normal eyelid conjunctiva were 23.53 and 8 / 34, respectively. The expression of PTEN protein in squamous cell carcinoma of eyelid conjunctiva was significantly lower than that in adjacent normal eyelid conjunctiva tissue (P 0.000). The positive expression rates of P0.01- ilk protein in conjunctival squamous cell carcinoma of eyelid and adjacent normal eyelid conjunctiva were 73.53 / 25 / 34 respectively. The expression of ilk protein in squamous cell carcinoma of eyelid conjunctiva was significantly higher than that in normal conjunctiva conjunctiva tissue adjacent to carcinoma, and the difference was significant. 2the expression of PTEN protein and ILK protein was not significantly different from that of age and sex. Ilk was found in 23 cases of well-differentiated conjunctival squamous cell carcinoma of eyelid. There were 14 cases of positive expression, all of which were positive in 11 cases of low differentiation group. There was a significant difference between the two groups (P < 0.017). In 23 cases of highly differentiated conjunctival squamous cell carcinoma of eyelid, 8 cases were positive, and all the cases were negative in 11 cases of low differentiation group. There was significant difference between the two groups. 3There were 8 cases of positive expression of PTEN protein in 34 cases of squamous cell carcinoma of eyelid conjunctiva, and 25 cases of positive expression of ilk protein in 26 cases of negative cases. The expression of PTEN protein and ILK protein was positive in 3 cases and negative in 6 cases in 9 cases. Of the 26 cases with negative expression of PTEN protein, 19 cases were positive for ilk protein expression, and 10 cases were positive for ilk protein expression. The expression of PTEN protein in 7 cases was positive, and Spearman grade correlation analysis showed that there was a negative correlation between them. Conclusion: 1: 1 PTEN? The expression of white in the adjacent normal eyelid conjunctiva tissue was higher than that in the squamous cell carcinoma of eyelid conjunctiva. The expression of PTEN protein was correlated with the degree of tumor differentiation. The results suggest that the low expression or absence of PTEN protein is associated with the development of squamous cell carcinoma of the eyelid conjunctiva. 2. The expression of ILK in the squamous cell carcinoma of eyelid conjunctiva was significantly higher than that in the adjacent normal eyelid conjunctival tissue. The results suggest that the expression of ILK protein may be related to the development of squamous cell carcinoma of eyelid conjunctiva. The results suggest that ILK may be related to the development and malignant potential of squamous cell carcinoma of eyelid conjunctiva. 3upregulation of ilk protein may inhibit the activity of PTEN and the deletion of PTEN protein may promote the expression of ILK protein. ILK gene may be a new target for the treatment of eyelid conjunctival squamous cell carcinoma.
