尼妥珠单抗对鼻咽癌CNE-2细胞作用的拉曼光谱研究

【摘要】:目的应用拉曼光谱技术研究尼妥珠单抗浓度和时间因素对鼻咽癌CNE-2细胞作用效果的影响。 方法培养并收集生长状态良好的人鼻咽癌CNE-2细胞,接种于四个细胞培养瓶中。向4个培养瓶中分别加入0,40ug/ml,150ug/ml,200ug/m四种浓度的尼妥珠单抗注射液各2ml,置于细胞培养箱中培养24h。采用Renishaw激光显微共焦拉曼光谱仪对四种药物浓度培养的CNE-2细胞进行测量。每个药物浓度组测量20个细胞,将测得的光谱进行PCA-LDA统计学分析,寻找最佳药物作用浓度。再将CNE-2细胞接种于另外三个培养瓶中,分别加入最佳药物浓度的尼妥珠单抗注射液各2ml,置于细胞培养箱中分别培养24h、48h、72h后进行拉曼光谱测量。 结果尼妥珠单抗注射液作用24h后,40ug/ml组的CNE-2细胞平均拉曼光谱与对照组无明显差别,150ug/ml组部分谱峰出现差别,200ug/ml组多个谱峰出现明显差别。经过PCA-LDA统计分析,随着药物浓度的提高,药物组与对照组的分离趋势越发明显,200ug/ml时,药物组和对照组能够完全区分。40ug/ml,150ug/ml,200ug/ml时判别的灵敏度分别为60%、85%、100%;特异度分别为70%、70%和100%。200ug/ml药物浓度下,24h组、48h组、72h组的拉曼光谱表现出明显的差异,但并未表现出与作用时间相关的变化趋势 结论拉曼光谱分析表明,在24h作用时间下,尼妥珠单抗对CNE-2细胞的作用效果随着药物浓度增加而增强,200ug/ml时的作用效果最为显著。在相同药物浓度下,药物作用效果与时间梯度未表现出明显的相关关系,可能与谱峰间的相互干扰有关,有待方法学的改进加以明确。
[Abstract]:Objective to investigate the effects of concentration and time of Nitozhu-McAb on nasopharyngeal carcinoma (NPC) CNE-2 cells by Raman spectroscopy. Methods Human nasopharyngeal carcinoma (NPC) CNE-2 cells in good growth state were cultured and inoculated into four culture flasks. Each of the four concentrations of Nietozumab injection was 2ml / 200ugrm. Cultured in cell incubator for 24 hours, CNE-2 cells cultured with four drug concentrations were measured by Renishaw laser microconfocal Raman spectrometer. 20 thin cells were measured in each drug concentration group. The cypress. The spectrum was analyzed by PCA-LDA statistics to find the best drug concentration. Then the CNE-2 cells were inoculated into the other three culture flasks. The optimal concentration of Nietozumab injection was 2 ml each and cultured in a cell incubator for 24 h or 48 h for 72 h. The Raman spectra were measured. Results there was no significant difference in the average Raman spectra of CNE-2 cells between the 40 g / ml group and the control group after 24 hours of treatment with Niytozumab injection. There was no significant difference in some peaks between the 150ug-ml group and the control group. After PCA-LDA statistical analysis, with the increase of drug concentration, the separation trend between the drug group and the control group became more and more obvious. At 200ugrml, the sensitivity of the drug group and the control group was 60% or 85%, respectively. Under the same drug concentration, there was no obvious correlation between the effect and the time gradient, and the effect might be related to the interference between the spectrum peaks. Improvements in methodology remain to be clarified.
