PPAR-γ在甲状腺相关性眼病患者眶脂肪和血清中的表达研究

【摘要】:目的:检测TAO患者眼眶脂肪组织以及血清中PPAR-γ的表达水平,初步探讨PPAR-γ在TAO发病过程中的作用。方法:选取TAO患者4例,健康对照者4例,用RT-PCR法检测两组眶脂肪PPAR-γmRNA的表达水平以及用Western blot法检测两组眶脂肪PPAR-γ蛋白的表达水平。选取TAO患者33例(男14例,女19例),按临床活动性评分(CAS)及99Tcm-DTPA眼眶显像将TAO患者分为两大组:活动期(CAS≥3,99Tcm-DTPA眼眶显像阳性)18例(男7例,女11例)和非活动期(CAS3,99Tcm-DTPA眼眶显像阴性)15例(男7例,女8例)。单纯Graves病(GD)患者16例(男6例,女10例),健康对照者(CON)15例(男7例,女8例)。用Elisa法检测四组血清PPAR-γ表达水平。采用SPSS19.0进行统计分析,计量资料采用x±s表示,计数资料采用例数(构成比)表示,正态分布数据均数的比较采用方差分析,非正态分布数据均数的比较采用秩和检验,构成比的比较采用卡方检验,检验水准α=0.05。结果:1.两组患者眶脂肪中PPAR-γmRNA的表达水平:TAO组2.640±0.525,CON组1.182±0.272,TAO组明显高于CON组,差异有统计学意义(P0.05)。2.眶脂肪中两组患者PPAR-γ蛋白的表达水平:TAO组0.992±0.104,CON组0.634±0.0291,T AO组明显高于CON组,差异有统计学意义(P0.05)。3.血清中四组患者PPAR-γ的表达水平:活动组0.0133±0.0031pg/ml,非活动组0.00903±0.00421bpg/ml,GD组0.0190±0.00323pg/ml,CON组0.00813±0.00774 pg/ml。四组比较,差异有统计学意义(F=16.199,P0.001),其中活动组与非活动组比较,活动组明显高于非活动组,差异有统计学意义(P=0.0150.05);活动组与GD组比较,活动组明显低于GD组,差异有统计学意义(P=0.001);活动组与CON组比较,活动组明显高于CON组,差异有统计学意义(P=0.0030.05);非活动组与GD组比较,非活动组明显低于G D组,差异有统计学意义(P0.001);非活动组与CON组比较,差异无统计学意义(P=0.6100.05);GD组与CON组比较,GD组明显高于CON组,差异有统计学意义(P0.001)。男女患者血清中PPAR-γ表达水平无统计学差异(P0.05),不同年龄患者血清中PPAR-γ表达水平亦无统计学差异(P0.05)。结论:1.TAO患者眶脂肪PPAR-γ基因转录水平及蛋白表达水平均显著增加。2.活动期TAO患者血清中PPAR-γ表达水平显著增高。3.甲状腺功能亢进患者血清中PPAR-γ表达水平显著增高。4.年龄和性别对TAO患者血清中PPAR-γ的表达无显著影响。
[Abstract]:Objective: to detect the expression of PPAR- 纬 in orbital adipose tissue and serum of TAO patients, and to explore the role of PPAR- 纬 in the pathogenesis of TAO. The expression of PPAR- 纬 mRNA in orbital fat and the expression of PPAR- 纬 protein in orbital fat were detected by RT-PCR method and Western blot method respectively. 19 female patients were divided into two groups according to the clinical activity score (CAS) and 99Tcm-DTPA orbital imaging: 18 cases (male 7 cases, female 11 cases) with CAS399Tcm-DTPA orbital imaging positive and 15 cases (male 7 cases) with CAS399Tcm-DTPA negative orbital imaging (male 7 cases), with CAS399Tcm-DTPA imaging negative (7 cases), the patients with CAS399Tcm-DTPA orbital imaging were divided into two groups: CAS399Tcm-DTPA imaging was positive in 18 cases (male 7 cases, female 11 cases). There were 16 patients (6 males, 10 females) with Graves disease and 15 healthy controls (7 males and 8 females). The expression of PPAR- 纬 in serum of the four groups was detected by Elisa method. SPSS19.0 was used for statistical analysis, and the measured data were expressed as x 卤s. The counting data are expressed by examples (composition ratio), the mean of normal distribution data is compared by ANOVA, the non-normal distribution data mean is compared by rank sum test, and the composition ratio is compared by chi-square test. Results the expression level of PPAR- 纬 mRNA in the orbital fat of the two groups was significantly higher than that in the CON group (2.640 卤0.525) and 1.182 卤0.272 respectively. The expression level of PPAR- 纬 protein in the two groups was significantly higher than that in the CON group (0.634 卤0.0291T AO, 0.992 卤0.104CON, 0.634 卤0.0291T). The expression level of PPAR- 纬 in the four groups was 0.0133 卤0.0031 PG / ml in the active group, 0.00903 卤0.00421 bpg / ml in the inactive group, 0.0190 卤0.00323pg / ml / ml group in the GD group, 0.00813 卤0.00774 pg / ml / ml in the four groups. There was significant difference in the expression of PPAR- 纬 between the active group and the inactive group, and the difference between the active group and the inactive group was significantly higher than that in the inactive group, and the expression of PPAR- 纬 in the active group was significantly higher than that in the inactive group. The difference between active group and GD group was significantly lower than that in GD group, the difference was statistically significant, and the difference between active group and CON group was significantly higher than that in CON group (P 0.0030.05), the difference between active group and GD group was statistically significant, and the difference between inactive group and GD group was significantly higher than that in active group and GD group. There was no significant difference between the inactive group and the CON group, and the difference between the GD group and the CON group was higher than that in the CON group. There was no significant difference in the expression of PPAR- 纬 between male and female patients and there was no significant difference in the expression of PPAR- 纬 between male and female patients with different ages. Conclusion the transcription level and protein expression of PPAR- 纬 gene in orbital fat of patients with TAO are not significantly different from those of patients of different ages. The expression of PPAR- 纬 in serum of patients with hyperthyroidism was significantly higher than that of patients with hyperthyroidism. 4. Age and sex had no significant effect on the expression of PPAR- 纬 in serum of patients with TAO.
