Skp2,p27,PTEN在眼部淋巴组织病变中的表达和意义

【摘要】:目的:探讨眼部淋巴组织病变中Skp2,p27和PTEN表达的意义以及三者表达的相关性,探索三者在眼部B细胞非霍杰金氏淋巴瘤表达的特点。 方法:收集青岛大学附属医院从1995年到2011年手术切除眼部B细胞非霍杰金氏淋巴瘤30例,反应性淋巴组织病变增生10例作为阳性对照组,淋巴瘤组按病理类型分类(根据2001年世界卫生组织修正的欧美淋巴瘤分类),取其中位于眼科前三位的B细胞NHL淋巴瘤类型,其中黏膜相关淋巴组织结外边缘区B细胞淋巴瘤19例,浆细胞瘤6例,弥漫大B细胞淋巴瘤5例,用免疫组化法分别检测标本中Skp2,p27禾PPTEN在淋巴瘤和反应性淋巴组织病变增生组的表达,并对两组的表达情况做对比,以病理分级作为淋巴瘤的独立因素进行分析。 结果:眼部B细胞非霍杰金氏淋巴瘤Skp2表达率与反应性淋巴组织增生相比显著增高(P0.05).淋巴瘤p27kipl,PTEN表达率与反应性淋巴组织增生相比显著降低(P0.05,P0.05)。三种蛋白的表达与患者的年龄,性别,发病部位无明显相关性(P0.05)。在淋巴瘤组中,Skp2在MALT,PL,DLBCL细胞核中的表达率是68.4%,83.3%,100%,p27kipl分别是84.2%,66.7%,40%,PTEN在三种淋巴瘤细胞质中的表达率分别是73.7%,33.3%,20%。随淋巴瘤病理分级的提高,Skp2在MALT, PL, DLBCL中的表达显著增高,而p27kipl和PTEN的表达显著降低。在MALT淋巴瘤中,我们发现Skp2与p27kipl呈负相关(r=—0.134,X2=12.58,p0.05),Skp2与PTEN呈负相关(r=-0.883,X2=20.52,p0.05)。而p27kipl与PTEN(r=0.828,X2=20.66,p0.05)呈正相关。 结论:眼部B细胞非霍杰金氏淋巴瘤中,Skp2的表达水平随病理分级恶性度的增加而逐渐增加,p27kipl,PTEN的表达水平水平随病理分级恶性度的增加而逐渐降低,Skp2高表达,p27kipl和PTEN(?)氐表达分别在眼部B细胞非霍杰金氏淋巴瘤的发生,发展中可能起一定的作用,在黏膜相关淋巴组织结外B细胞淋巴瘤中,Skp2与p27kipl,PTEN的表达水平呈负相关,p27kipl与PTEN呈正相关。联合三种蛋白的表达可以作为检测眼部B细胞非霍杰金氏淋巴瘤的指标。
[Abstract]:Objective: to investigate the significance of Skp2p27 and PTEN expression in ocular lymphoid tissue lesions and the correlation between the expression of Skp2p27 and PTEN, and to explore the expression characteristics of Skp2p27 and PTEN in ocular B cell non-Hodgkin 's lymphoma. Methods: from 1995 to 2011, 30 cases of ocular B-cell non-Hodgkin 's lymphoma and 10 cases of reactive lymphoid tissue hyperplasia were collected from Qingdao University affiliated Hospital as positive control group. The lymphoma group was classified according to pathological types (according to the classification of lymphoma in Europe and the United States as amended by the World Health Organization in 2001), B-cell NHL lymphoma, which was located in the top three in ophthalmology, was selected. There were 19 cases of B cell lymphoma, 6 cases of plasmacytoma and 5 cases of diffuse large B cell lymphoma. The expression of Skp2p27 and PPTEN in lymphoma and reactive lymphoid tissue hyperplasia was detected by immunohistochemical method. The expression of Skp2p27 and PPTEN was compared between the two groups, and the pathological grade was used as an independent factor to analyze the lymphoma. Results: the expression rate of Skp2 in ocular B cell non-Hodgkin 's lymphoma was significantly higher than that in reactive lymphoid tissue proliferation (P 0.05). The expression rate of p27kipline PTEN was significantly lower than that of reactive lymphoid tissue proliferation. The expression of three proteins and the age of the patients were significantly lower than those of reactive lymphoid tissue proliferation. Sex, In the lymphoma group, the expression rate of Skp2 in the DLBCL cell nucleus was 68.4%. The expression rate of Skp2 in the cytoplasm of the three types of lymphoma was 73.733. 3. The expression rate of Skp2 in MALTPLPLDLBCL was significantly higher than that of the control group, and the expression of Skp2 in MALTPLPLDLBCL was significantly increased with the increase of pathological grade of the lymphoma, and the expression rate of PTEN in the cytoplasm of the three kinds of lymphoma was 73.733. 3%, respectively, and the expression rate of Skp2 in MALTPLDLBCL was significantly higher than that in the control group. However, the expression of p27kipl and PTEN was significantly decreased in MALT lymphoma. In MALT lymphoma, we found that there was a negative correlation between Skp2 and p27kipl. It was found that there was a negative correlation between Skp2 and p27kipl. There was a positive correlation between p27kipl and p27kipl, p0.05kip2 and PTEN, and a positive correlation between p27kipl and p27kipl and p27kipl and p27kipl were positively correlated with p27kipl and p27kipl, and between p27kipl and p27kipl were negatively correlated with p27kipl and p27kipl, respectively. Conclusion: the expression level of Skp2 in ocular B cell non-Hodgkin 's lymphoma increases gradually with the increase of pathological grade malignancy. The expression level of p27kipl and PTEN decreases with the increase of pathological grade malignancy. ) Di expression may play a certain role in the development and development of ocular B-cell non-Hodgkin 's lymphoma. There was a negative correlation between p27kipl and PTEN in the expression of Skp2 and p27kipl in extranodular B cell lymphoma of mucosa-associated lymphoid tissue. The expression of three proteins combined with these proteins could be used as a marker for detecting ocular B cell non-Hodgkin 's lymphoma.
