Akt促进大鼠角膜新生血管形成的初步研究

【摘要】:目的观察大鼠角膜碱烧伤新生血管模型中蛋白激酶B(Akt)的表达,探讨Akt在角膜新生血管形成过程中的作用。 方法将20只成年大鼠分为正常对照组A组(5只5眼)及实验组(15只15眼),实验组按角膜碱烧伤后的不同时间分为B、C、D三组,每组各5只5眼。用浸泡1%NaOH溶液的直径3mm大小圆形滤纸贴敷于实验组大鼠的右眼角膜中央40s,B组为角膜碱烧伤后3d,C组为角膜碱烧伤后5d,D组为角膜碱烧伤后14d。自角膜碱烧伤24小时后开始每日行裂隙灯检查,观察各组角膜新生血管生长情况,并用western印迹法检测各组角膜组织中Akt表达的变化。 结果1.在大鼠角膜碱烧伤后24小时,角巩缘血管开始扩张,48小时角巩缘处开始有血管向透明角膜生长,第14天角膜遍布新生血管,血管长而稀疏,角膜中央变薄。2.在大鼠角膜碱烧伤新生血管模型中,C组Akt蛋白表达水平最高,为0.840±0.015,与其他各组相比Akt蛋白表达差异具有统计学意义(P0.05),各实验组与正常对照组相比Akt蛋白表达差异具有统计学意义(P0.05)。 结论1.在大鼠角膜碱烧伤后一定时间内,Akt的表达增强并呈上升趋势。2.Akt在大鼠角膜碱烧伤新生血管形成过程中可能具有促进作用。
[Abstract]:Objective To observe the expression of protein kinase B (Akt) in the neovascularization model of rat corneal alkali burn, and to explore the role of Akt in the formation of corneal neovascularization.
Methods 20 adult rats were divided into normal control group and A group (5 eyes of 5) and experimental group (15 eyes of 15), the experimental group at different time after corneal alkali burn were divided into B, C, D three groups, 5 rats in each group. 5 eyes with 3mm diameter size of circular filter paper soaked 1%NaOH solution is applied to the experimental group rats right central corneal 40s, B group after corneal alkali burn 3D group C after corneal alkali burn 5D group D after corneal alkali burn 14D. corneal alkali burn for 24 hours after the daily slit lamp examination, to observe the growth of corneal neovascularization. Change and Western blotting to detect the expression of Akt in corneal tissue.
Results 1. corneal alkali burn in rats after 24 hours, the limbus vessels began to expand, 48 hours perilimbal began to clear corneal vascular growth, fourteenth days in corneal neovascularization, vascular and sparse, central corneal thinning.2. in corneal neovascularization after corneal alkali burn model of rats in C group, Akt the expression of the highest level, 0.840 + 0.015, compared with the other groups were statistically significant differences in expression of Akt protein (P0.05), the experimental group compared with the normal control group was statistically significant difference in Akt protein expression (P0.05).
Conclusion 1., the expression of Akt increased and increased in a certain time after corneal alkali burn in rats..2.Akt may play a role in promoting angiogenesis in rat corneal alkali burn.
