LXR在湿性AMD患者中的表达及与血脂代谢的相关性分析

【摘要】:目的:探索肝脏X受体(liver X receptor,LXR)在湿性年龄相关性黄斑变性(age-related macular degeneration,AMD)患者外周血单个核细胞(peripheral blood mononuclear cells,PBMCs)的表达及与血脂代谢的相关性分析。方法:随机选取本院眼科门诊湿性AMD患者20例,同时选取健康志愿者20例作为对照组。收集年龄、性别等病史资料,通过抽血测定血清总胆固醇(total cholesterol,TC)、甘油三酯(triglyceride,TG)、高密度脂蛋白胆固醇(high density lipoprotein cholesterol,HDL)、低密度脂蛋白胆固醇(low density lipoprotein cholesterol,LDL)水平。采用荧光定量PCR(q RT-PCR)的方法检测LXRα和LXRβ的m RNA的表达水平。使用LXR激动剂TO901317处理,观察PBMC中LXRα和LXRβ的变化。采用t检验、非参数检验、Pearson相关性分析及ROC分析进行数据统计分析。结果:湿性AMD患者中LXRα,LXRβ的m RNA的表达水平都低于对照组的表达水平(t=3.881,P=0.001;t=5.839,P=0.000)。使用LXR激动剂TO901317处理后,在PBMC中LXRα的m RNA表达水平升高,LXRβ表达无差异。Pearson相关性分析提示LXRα与TG具有负性相关性,而LXRα与TC、HDL、LDL及LXRβ与TC、TG、HDL、LDL均无相关性。ROC分析得LXRα检测结果的曲线下面积(AUC)为0.824(95%CI=0.676~0.973,P=0.001),LXRβ检测结果的AUC为0.945(95%CI=0.873~1.000,P=0.000)。结论:LXRα和LXRβ的表达水平下降提示与湿性AMD疾病的发生发展密切相关,可能成为诊断和治疗湿性AMD的一个新靶点。
[Abstract]:Objective: to investigate the expression of liver X receptor blood X receptor (LXR) in peripheral blood mononuclear cells (PBMC) from patients with age-related macular degenerationin patients with wet age-related macular degeneration and its correlation with blood lipid metabolism. Methods: ophthalmic departments were randomly selected from our hospital. 20 cases of AMD were diagnosed. At the same time, 20 healthy volunteers were selected as control group. The expression of m RNA of LXR 伪 and LXR 尾 was detected by taking blood samples from serum total cholesterol cholesterol, triglyceride triglyceride, high density lipoprotein cholesterol, low density lipoprotein cholesterol low density lipoprotein cholesterol cholesterol (LDL). The expression of LXR 伪 and LXR 尾 was detected by fluorescence quantitative PCR(q RT-PCR. Level. Treated with LXR agonist TO901317, The changes of LXR 伪 and LXR 尾 in PBMC were observed. Results: the expression level of m RNA of LXR 伪 -LXR 尾 in patients with wet AMD was lower than that of control group (P < 0.05). After treated with LXR agonist TO901317, the expression level of LXR 伪 -LXR 尾 was significantly lower than that of control group. There was no difference in m RNA expression of LXR 伪 in PBMC. Pearson correlation analysis showed that there was a negative correlation between LXR 伪 and TG. However, there was no correlation between LXR 伪 and LXR 尾 and between LXR 尾 and LXR 尾. The area under the curve of LXR 伪 detection results was 0.82495 CI 0.676 ~ 0.973C ~ (0.001) AUC was 0.945 95CI0.8731.000 ~ 0.0000.0000.Conclusion the decrease of the expression levels of LXR 伪 and LXR 尾 is closely related to the occurrence and development of the disease.
