翼状胬肉和球结膜中增殖和凋亡相关因子表达情况的研究

【摘要】:目的:研究翼状胬肉和球结膜组织中增殖和凋亡相关因子的表达情况,探讨增殖和凋亡的平衡在翼状胬肉发病机制所起到的作用,为临床上探寻翼状胬肉的非手术治疗方法提供理论依据。 方法:临床收集手术切除的翼状胬肉组织标本33例,收集正常球结膜组织23例作为对照。经过福尔马林固定,石蜡包埋,切片。HE染色,免疫组织化学方法检测翼状胬肉和球结膜组织中PCNA、Ki-67、Bax、Bcl-2、mP53、Caspase-3的表达情况,并使用TUNEL染色法检测翼状胬肉和正常球结膜组织中处于凋亡的细胞。并进行统计学分析比较。 结果: PCNA在33例翼状胬肉组织样本中24例阳性,23例球结膜组织中7例阳性(P0.01);Ki-67在翼状胬肉和球结膜组织中均有表达,但是在翼状胬肉组织中阳性细胞数平均为14.27±4.43,球结膜组织总阳性细胞数平均为4.26±2.42(P0.01);mP53在33例翼状胬肉组织样本中15例阳性,在球结膜组织中全部为阴性(P0.01);Bcl-2在33例翼状胬肉组织样本中全为阳性,在23例球结膜组织中2例阳性(P0.00);Bax在翼状胬肉和球结膜组织中均有表达;Caspase-3在33例翼状胬肉组织标本中14例阳性,在23例球结膜组织中13例阳性,差异无统计学意义(P=0.299);TUNEL染色显示,翼状胬肉组织中凋亡细胞局限在上皮基底层细胞,而球结膜组织中整个上皮层均存在凋亡细胞。 结论:细胞增殖和细胞凋亡均参与翼状胬肉发病,翼状胬肉中细胞增殖明显增强,细胞凋亡明显减弱,细胞增殖和凋亡之间平衡的打破导致翼状胬肉的发病。
[Abstract]:Objective: to study the expression of proliferative and apoptosis-related factors in pterygium and bulbar conjunctiva, and to explore the role of the balance of proliferation and apoptosis in the pathogenesis of pterygium. To provide theoretical basis for exploring non-operative treatment of pterygium. Methods: 33 specimens of pterygium were collected and 23 cases of normal bulbar conjunctiva were collected as control. The specimens were fixed with formalin, embedded in paraffin, stained with HE. The expression of Bcl-2mP53Caspase-3 in pterygium and bulbar conjunctiva was detected by immunohistochemical method, and the apoptotic cells in pterygium and normal bulbar conjunctiva were detected by TUNEL staining. Results: PCNA was expressed in pterygium and bulbar conjunctiva in 24 out of 33 pterygium tissue samples. However, the average number of positive cells in pterygium was 14.27 卤4.43 and the total positive cell count in bulbar conjunctiva was 4.26 卤2.42 P0.01mp53 in 33 pterygium tissues. In the bulbar conjunctival tissue, all the specimens of p0.01 and Bcl-2 were positive in 33 cases of pterygium. The expression of Caspase-3 in pterygium and bulbar conjunctiva was positive in 2 out of 23 cases of bulbar conjunctiva and in 33 cases of pterygium and 13 cases in 23 cases of bulbar conjunctiva. The apoptotic cells in pterygium were confined to the basal layer of epithelium, while apoptotic cells were found in the whole epithelium of bulbar conjunctiva. Conclusion: cell proliferation and apoptosis are involved in the pathogenesis of pterygium. The proliferation and apoptosis of pterygium are significantly increased. The imbalance between cell proliferation and apoptosis leads to the pathogenesis of pterygium.
