Snail、E-cadherin、VEGF蛋白及其mRNA在喉鳞癌中的表达研究

【摘要】:本课题通过应用EliVisionTM Plus免疫组织化学技术(Immunohistochemistry, IHC)(?)原位分子杂交技术(In situ hybridization,ISH)分别检测喉黏膜慢性炎、不典型增生、鳞癌(Laryngeal squamous cell carcinoma, LSCC)组织中Snail、E-cadherin和VEGF蛋白及其mRNA的表达情况。 探讨Snail、E-cadherin和VEGF蛋白及其mRNA分别在喉黏膜慢性炎、不典型增生和鳞癌组织中的表达及与患者临床病理特征的关系;探讨Snail、E-cadherin和VEGF蛋白在喉鳞癌组织中表达的相互关系;探讨Snail、E-cadherin和VEGF mRNA在喉鳞癌组织中表达的相互关系;免疫组织化学技术和原位分子杂交技术两种方法检测喉鳞癌组织中Snail、E-cadherin和VEGF表达的一致性分析。 收集解放军第251医院病理科2005年1月至2007年1月间的喉鳞癌术后标本60例、不典型增生30例和黏膜慢性炎组织20例,分别采用EliVisionTM Plus免疫组织化学技术和原位分子杂交技术检测Snail、E-cadherin和VEGF蛋白及其mRNA的表达情况,并分析它们与患者的性别、年龄、病理分级、TNM分期、T分期、甲状软骨累及、淋巴结转移及远处转移的关系;分析两种方法检测Snail、E-cadherin和VEGF表达的一致性。 Snail、E-cadherin和VEGF蛋白及其mRNA在60例喉鳞癌组织中的阳性率分别为:78.3%(47/60)、46.7%(28/60)、80.0%(48/60)及70.0%(42/60)、45.0%(27/60)、73.3%(44/60);在30例喉黏膜不典型增生组织中的阳性率分别为:43.3%(13/30)、76.7%(23/30)、36.7%(11/30)及20.0%(6/30)、70.0%(21/30)、26.7%(8/30);在20例喉黏膜慢性炎组织中的阳性率分别为:15.0%(3/20)、100%(20/20)、15.0%(3/20)及5.0%(1/20)、90.0%(18/20)、5.0%(1/20)。Snail、VEGF蛋白及其mRNA在喉鳞癌组织的阳性率显著高于不典型增生和黏膜慢性炎组织的阳性率,E-cadherin蛋白及其mRNA在喉鳞癌组织的阳性率均显著低于不典型增生和黏膜慢性炎组织的阳性率,差异具有统计学意义。不典型增生组织中Snail蛋白阳性率高于黏膜慢性炎组织的表达率,差异有统计学意义,Snail mRNA、 VEGF蛋白及其mRNA阳性率高于黏膜慢性炎组织的表达率,但差异无统计学意义。不典型增生组织中E-cadherin蛋白阳性率低于黏膜慢性炎组织的表达率,差异具有统计学意义,E-cadherin mRNA阳性率低于黏膜慢性炎组织的表达率,但差异无统计学意义;Snail、 VEGF蛋白及mRNA在喉鳞癌组织中的高表达及其E-cadherin蛋白及mRNA的低表达与肿瘤TNM分期、T分期、甲状软骨侵犯、淋巴结转移、远处转移及5年生存率密切相关,而与年龄、性别无明显关系;Snail蛋白表达与病理分级有关,而Snail mRNA的表达与病理分级无关。 60例喉鳞癌组织中Snail、E-cadherin、 VEGF蛋白及mRNA阳性表达分别为47例、28例、48例及42例、27例、44例,经Spearman相关分析,结果显示:Snail蛋白及mRNA与E-cadherin蛋白及mRNA表达呈显著负相关;Snail蛋白及mRNA与VEGF蛋白及mRNA表达呈显著正相关;E-cadherin蛋白及mRNA与VEGF蛋白及mRNA表达呈显著负相关。 Snail过表达与喉鳞癌的发生有密切关系,它可能参与了癌前细胞向癌细胞转化的调控机制;Snail、 E-cadherin和VEGF在免疫组化和原位杂交两种检测方法中具有一致性;Snail、 E-cadherin和VEGF共同参与了喉鳞癌的发生、发展、浸润、转移过程,是喉鳞癌发生浸润和转移的重要生物学指标,联合检测Snail、 E-cadherin和VEGF对预测肿瘤转移和预后有重要意义,Snail将有可能成为喉鳞癌靶向治疗的一个新指标。
[Abstract]:This topic through the application of EliVisionTM Plus immunohistochemical technique (Immunohistochemistry, IHC) (?) in situ hybridization (In situ hybridization, ISH) were detected in laryngeal mucosa of chronic inflammation, atypical hyperplasia, squamous cell carcinoma (Laryngeal squamous cell carcinoma, LSCC) in Snail, the expression of E-cadherin and VEGF protein and mRNA.
The effects of Snail, E-cadherin and VEGF protein and mRNA respectively in laryngeal mucosa of chronic inflammation, the expression of atypical hyperplasia and squamous cell carcinoma and its relationship with the clinical pathological features of patients; to investigate the Snail, relationship between expression of E-cadherin and VEGF protein in laryngeal squamous cell carcinoma; to explore the relationship between the expression of Snail, E-cadherin and VEGF mRNA in laryngeal squamous cell carcinoma the organization of Snail in laryngeal squamous cell carcinoma; immunohistochemistry and in situ hybridization of two kinds of detection methods, consistency analysis of the expression of E-cadherin and VEGF.
60 samples were collected from the 251st Hospital of PLA from January 2005 to January 2007 in the Department of pathology of postoperative laryngeal squamous cell carcinoma, 30 cases of atypical hyperplasia and chronic inflammation of mucosa tissues in 20 cases were measured by Snail EliVisionTM Plus immunohistochemistry and in situ hybridization, the expression of E-cadherin and VEGF protein and mRNA, and analyze them with patients the gender, age, pathological grading, TNM staging, T staging, thyroid cartilage involvement, lymph node metastasis and distant metastasis; analysis of two methods for detection of Snail, E-cadherin and VEGF expression of consistency.
