维吾尔族、汉族变应性鼻炎患者IL-4、IFN-γ基因甲基化的研究

【摘要】:目的:针对维吾尔族、汉族变应性鼻炎患者和正常健康人IL-4、IFN-γ基因甲基化进行研究,分别探讨IL-4、IFN-γ基因甲基化在维吾尔族、汉族AR患者与健康者是否存在差异性。方法:选择维吾尔族、汉族AR患者各50例,维吾尔族、汉族正常人各50例,提取外周血液DNA,,运用甲基化特异性PCR(MSP)方法检测IL-4、IFN-γ基因甲基化。结果:汉族AR病例组与正常对照组IL-4基因甲基化率的比较具有统计学意义(P0.05)。汉族与维族正常对照组之间IL-4基因甲基化率的比较有统计学意义(P0.05)。汉族与维族AR病例组IL-4基因甲基化率的比较,无统计学意义(P0.05)。维族AR病例组与正常对照组IL-4基因甲基化率的比较,无统计学意义(P0.05)。汉族、维族AR病例组以及AR病例组与正常对照IFN-γ基因甲基化率的比较,均无统计学意义(P0.05),汉族与维族正常对照组IFN-γ基因甲基化率的比较有统计学意义(P0.05),维、汉族变应性鼻炎患者皮肤变应原点刺实验中艾蒿和藜属阳性率最高,维、汉族相比差异无统计学意义(P0.05)。结论:艾蒿和藜属是维、汉族AR患者室外主要吸入性变应原。维吾尔族与汉族AR病例组之间IL-4基因启动子区甲基化阳性结果无差异,但汉族病例组与汉族健康对照组阳性结果有差异,说明IL-4基因甲基化可能在变应性鼻炎的发病过程中发挥着重要作用。汉族健康人群中IL-4基因半甲基化高于维吾尔族健康人群,说明汉族人群中的IL-4甲基化有可能趋向于阳性表达,汉族人群易患上变应性鼻炎。维吾尔族与汉族AR病例组以及正常对照组之间IFN-γ启动子区甲基化阳性结果均无差异性,但汉族与维族正常对照组之间有统计学差异,IFN-γ基因甲基化在变应性鼻炎作用有待于进一步研究。
[Abstract]:Objective: to study the methylation of IFN- 纬 gene in Uygur, Han nationality patients with allergic rhinitis and normal controls, and to explore the methylation of IFN- 纬 gene in Uygur nationality. Methods: there were 50 cases of AR in Uygur nationality, 50 cases in Han nationality, 50 cases in Uygur nationality and 50 cases in normal Han nationality. The DNA of peripheral blood was extracted, and the methylation of IFN- 纬 gene of IL-4 was detected by methylation specific PCR- MSP.Results: the methylation rate of IL-4 gene in AR patients of Han nationality was significantly higher than that in normal controls (P 0.05). The normal controls of Han nationality and Uygur nationality were also used to detect the methylation of IL-4 IFN- 纬 gene. The comparison of methylation rate of IL-4 gene between Han and Uygur patients was statistically significant (P 0.05), and the comparison of IL-4 methylation rate between Han and Uygur AR patients, There was no significant difference in methylation rate of IL-4 gene between Uygur and normal controls. The methylation rate of IFN- 纬 gene in Han, Uygur and AR cases and normal controls were compared. The methylation rate of IFN- 纬 gene in Han nationality and Uygur group was significantly higher than that in Uygur group. The positive rates of Artemisia annua and Chenopodium were the highest in the skin allergen prick test of patients with allergic rhinitis in Han nationality. Conclusion: Artemisia Artemisia and Chenopodium are the main outdoor inhalants in AR patients of Han nationality. There is no difference in methylation of promoter region of IL-4 gene between Uygur and Han patients. However, the positive results of IL-4 gene methylation may play an important role in the pathogenesis of allergic rhinitis in the Han nationality and the Han healthy control group, and the semi-methylation of the IL-4 gene in the Han nationality healthy population is higher than that in the Uygur healthy population, which indicates that methylation of IL-4 gene may play an important role in the pathogenesis of allergic rhinitis. The results indicated that IL-4 methylation might tend to positive expression in Han population, and the Han population was susceptible to allergic rhinitis. There was no difference in methylation positive results between Uygur and Han patients with AR and normal controls. However, there were significant differences between the Han and Uygur normal controls. The role of IFN- 纬 gene methylation in allergic rhinitis needs to be further studied.
